大鼠肢体缺血再灌注不同时段相关炎性介质的表达 被引量:8

Expression of correlated inflammation mediators on different time phases in rats with limbs ischemia-reperfusion

摘要 目的观察大鼠肢体缺血再灌注相关炎性因子随时间的延长而变化的情况及探讨其致远隔脏器损伤的意义。方法采用大鼠右下肢缺血再灌注模型,50只SD大鼠随机分成假手术组和缺血再灌注组(IR组),每组又分成再灌注3h、6h、9h、12h、15h各5个小组。ELISA测定各组血清IL-1β、IL-6,免疫组化检测肺组织TNF-α、MPO(髓过氧化物酶)、NF-κB的表达。结果与假手术组比较,IR组各时点血清IL-1β、IL-6及肺组织TNF-α、MPO、NF-κB的表达升高(P<0.05),并且各炎性因子随再灌注时间延长升高,IL-1β、IL-6、TNF-α、MPO在12h组达到高峰,NF-κB在9h组达到高峰(P<0.05);假手术组不同时段各炎性因子变化差异无显著性(P>0.05)。结论 IL-1β、IL-6、TNF-α、MPO及NF-κB参与了肢体缺血再灌注后肺损伤的发生机制。 Objective To investigate the changes of correlated inflammation factors as the time extends in rats with limbs ischemia - reperfusion and explore its significance on remote organs injury. Methods Adopting model of rats with right lower limb ischemia - reperfusion,50 Sprague- Dawley rats were randomly divided into the sham operated group and the ischemia- reperfusion group( IR group), each group was divided into 5 groups including 3 hours,6 hours,9 hours,12 hours and 15 hours after reperfusion. Expression of IL - 1β and IL -6 in blood serum were determined by ELISA, while TNF - α.MPO, NF - κB in lung tissue by immunohistochemistry. Results Compare with the sham group, Expression of IL - 1β and IL -6 in blood serum and TNF -α.MPO,NF - κB in lung tissue increased on different time phases in the IR group ( P 〈 0. 05 ), each factors in the IR group got a higher expression as time prolonged, with IL - 1β , IL - 6. TNF - α and MPO reached the peak value in 12 hours - group while NF - κB in 9 hours - group( P 〈 0.05). Expression of each each inflammation factor in the sham group had no significant change (P 〉 0.05 ). Conclusion IL - 1β, IL - 6. TNF - α, MPO and NF - κB may participate in the mechanism of lung injury after limbs ischemia - reperfusion.
