多囊肾小鼠肾功能的改变 被引量:6

摘要 目的探讨多囊肾脏组织特异性敲除小鼠(pkd1flox/-;Ksp-Cre mouse,简称PKD小鼠)肾功能在出生后随天数增加的变化情况。方法按SP级动物饲养标准进行饲养和繁殖PKD小鼠,选取出生后第1、3、6、91、2天五个时间点将PKD小鼠及野生小鼠(简写为WT组,下同)处死,分别检测其各时间点肾脏脏器系数,并应用尿素检测试剂盒对各时间点小鼠肾脏组织、血清中尿素含量进行检测。结果 PKD小鼠肾脏组织脏器系数在各时间点均明显高于野生组,P<0.05;PKD小鼠肾脏组织及血清中尿素含量在各时间点明显高于WT组,P<0.05;并随出生时间延长逐渐升高。结论本研究发现PKD小鼠出生后随着存活时间延长肾功能发生明显改变,并在生后两周内出现肾功能衰竭的相关症状。 Objective To approach postnatal polycystic kidney tissue specificity knockout mouse(pkd1flox/-;Ksp-Cre,below to abbreviate,PKD mause) mouse renal function variance condition.Methods We breed polycystic kidney tissue specificity knockout mouse(pkd1flox/-;Ksp-Cre,below to abbreviate PKD mouse) in our laboratory and select postnatal PKD and wild mouse in 1,3,6,9,12 day,than kill them and detect kidney coefficient.We detect urea content of kidney tissue,and serum to make use of urea detection kit on each time.Results The results display all PKD mouse's kidney coefficient on each time increased obviously than wild type(simplified character WT),P0.05;the urea content of PKD mouse kidney tissue and serum are higer than WT group(P0.05) and gradually advance to follow birth time extension.Conclusion In this study we discover that the renal function of postnatal PKD mouse changes obviously following the extension of times and PKD mouse appears renal failure symptoms in two weeks.
