摘要
目的:探讨含雌激素的冷冻保护剂对正常精液精子玻璃化冻存效果的影响。方法:将门诊收集的每例正常健康人精液平均分成5份,1份用于冷冻前指标测定,其余4份分别应用冷冻保护剂(CPM)甘油、甘油-卵黄-柠檬酸钠(GYC)、甘油+雌二醇(E2)、GYC+E2进行玻璃化冻存,检测冻存前、后精子活率、活力(a+b级)、正常精子形态百分率和精子尾部低渗肿胀率(HOSR)等精子功能的指标,评价4种CPM的冷冻保护效果。结果:4种CPM对精子的冷冻保护效果有区别,精子活率和活力、正常形态率和HOSR在冷冻后都有不同程度下降(P<0.01),甘油组、GYC组比甘油+E2组和GYC+E2组下降更明显(P<0.01)。GYC+E2组各参数高于甘油+E2组。结论:精子冷冻保护剂中加入雌激素可减少对正常精液冻融过程中精子的损伤,含雌激素的甘油复合型冷冻保护剂冷冻保护效果优于含雌激素的甘油型冷冻保护剂。
Objective: To explore the different effect of different cryoprotective containing estrogen on human spermatozoa.Methods: Each semen came from healthy patient will be divided into five copies,one copy was as control group,and the other four copies were as experimental group which were preserved by cryopreservativemediums(CPMs) with glycerol,glycerol-eygyolk-sodium(GYC),glycerol+estrogen and GYC+estrogen.The cryoprotective effects of human spermatozoa were evaluated by conventional semen analysis,viability,motility,morphology and spermatozoa hypo-osmotic swelling rate(HOSR).Results: During the procedure of cryoprotective and thawing,different CPM had different protective effect on the spermatozoa,which showed significantly different(P0.01),and glycerol and glycerol-eygyolk-sodium(GYC) group were decreased significantly than the other two groups(P0.01).The parameters in the the group of GYC+E2 were significantly higher than those of in the group of glycerol+E2(P0.01).Conclusions: The cryoprotectant containing estrogen can increase the function of cryopreserved sperm.
