加味犀角地黄汤对大鼠CIA急性关节肿胀与滑膜组织中VEGF表达的影响

Effect of added Xijiaodihuang decoction on VEGF level in joint synovial tissue s of CIA model rats

目的通过测定大鼠关节滑膜组织中血管内皮生长因子(VEGF)在大鼠胶原诱导性关节炎(CIA)模型形成及应用加味犀角地黄汤(XDD)治疗过程中的表达水平,探讨加味XDD对类风湿关节炎(RA)的治疗作用。方法用SD大鼠制作Ⅱ型胶原蛋白诱导的CIA大鼠模型,实时荧光定量PCR法检测正常组、模型组和实验组大鼠在第42d关节滑膜组织中VEGF的表达情况。结果模型组大鼠关节滑膜组织中VEGF的表达明显高于正常组,加味XDD治疗组VEGF的表达低于模型组。结论加味XDD可降低大鼠关节滑膜组织中VEGF的表达,可能会对RA急性期有一定的治疗作用。

Objective To explore the therapeutic effect of the Added Xijiaodihuang Decoction for rheumatoid arthritis by determining the levels of vascular endothelial growth factor（VEGF） in the joint synovial tissue of rats during the derivation of collagen-induced arthritis（CIA）.Methods SD rats were used to establish type II CIA rat model.VEGF levels were determined by real-time fluorescence quantitative RT-PCR at the 42th day after the induction in the rats of control,model and experimental group.Results VEGF level in the model group was obviously higher than that in control group,and the level in the experimental group was much lower than that in the model group.Conclusion Added Xijiaodihuang Decoction showed a role of reducing VEGF level in joint synovial tissue of CIA model rats and may have certain therapeutic effect for the acute stage of RA.
