摘要
采用复合酶酶解核桃蛋白,以水解度为考察指标,研究了底物质量浓度、复合酶配比、加酶量、pH、酶解温度、酶解时间对酶解的影响,并采用响应面法优化了Alcalase2.4L与胰蛋白酶复合酶解核桃蛋白的工艺参数。最佳酶解条件为:底物质量浓度40 g/L,Alcalase2.4L与胰蛋白酶的复合配比3∶1,pH7.4,酶解温度60.46℃,加酶量5.71%,酶解时间5.05 h。最佳条件下核桃蛋白水解度可达14.54%。
The degree of hydrolysis (DH) was used as an index, and the preparation of walnut peptides was studied by hydrolyzing walnut protein with compound enzyme of Alcalase2.4L and trypsin. By single factor tests and quadratic regression orthogonal rotation combination general design, the hydrolysis condition model was established. The results showed that the DH reached 14.54% under the optimum hydrolysis conditions of concentration of substrate 40 g/L , the proportion of compound enzyme (Alcalase2. 4L and trypsin) 3: 1, pH 7.4 ,temperature 60.46℃, enzyme dosage 5.71% and time 5.05 h.
