何首乌含药血清促进MSCs增殖的效应及机理研究 被引量:12

In vitro Effect of Radix Polygoni Multiflori Medicated Serum on Promoting MSCs Proliferation and Its Mechanism

摘要 目的探讨何首乌含药血清体外对骨髓间充质干细胞(MSCs)增殖的影响及机理研究。方法分离、培养大鼠MSCs,何首乌含药血清诱导培养MSCs 72h后,MTT法检测大鼠MSCs增殖情况;RT-PCR检测增殖过程中不同细胞因子mRNA表达;Real-time PCR检测SCF mRNA表达;Western blot检测SCF蛋白表达;ELISA法检测培养上清液SCF含量。结果 10%何首乌含药血清具有明显促进MSCs增殖的作用,并明显促进SCF mRNA表达及可溶性SCF蛋白的分泌。结论 10%何首乌含药血清具有促进MSCs增殖的作用,可能与其促进SCF mRNA表达、诱导MSCs分泌可溶性SCF蛋白增多有关。 Objective To investigate the in vitro effect of Radix Polygoni Multiflori medicated serum on bone marrow mesenchymal stem cells (MSCs)proliferation and to explore its mechanism. Methods After culturing MSCs with Radix Polygoni Multiflori medicated serum for 72 hours, the proliferation of MSCs was detected by MTT, cytokine mRNA expression by RT-PCR, stem cell factors (SCF)mRNA expression by Real-time PCR, SCF protein expression by Western blot, and SCF content of supernatant by ELISA assay. Results 10 % Radix Polygoni Multiflori medicated serum had stronger effect than the blank serum group on promoting MSCs proliferation, SCF mRNA expression and soluble SCF protein secretion. Conclusion 10 % Radix Polygoni Muhiflori medicated serum can promote the proliferation of MSCs, and the mechanism involves the induction of SCF mRNA expression and the increase of soluble SCF protein.
