脑忆源片对大鼠脑组织中5种单胺类递质含量的影响 被引量:4

Effect of Naoyiyuan Tablets on Contents of Five Monoamine Neurotransmitters in Rat Cerebral Homogenate

摘要 目的研究脑忆源片对大鼠脑组织中单胺类递质含量的影响,为探讨其改善学习记忆作用的机制提供理论依据。方法大鼠连续给药15 d后取样,采用HPLC-ECD法检测空白组和给药组大鼠脑组织中单胺类神经递质的含量;色谱柱:Phenomenex luna C18(150×4.60 mm,5μm);流动相:200 mmol.L-1磷酸二氢钠水溶液-乙腈(87∶13);流速:1 ml.min-1;柱温:35℃。结果 NE、E、DA、5-HIAA及5-HT五种物质在0.0095~10.5 ng.μL-1之间线性关系均良好(r≥0.9996);E的最低检测限低于5 ng.mL-1,NE、DA、5-HIAA、5-HT的最低检测限均低于1.21 ng.mL-1,五种物质平均回收率均在84.41%~100.92%之间。与空白组相比,高、中、低剂量组NE、E、DA含量均升高,5-HT含量均降低,且NE、DA及5-HT含量变化与剂量呈量效关系。结论该方法具有准确、灵敏、简便等优点,可用于大鼠脑组织中单胺类神经递质的检测;脑忆源片能明显提高大鼠脑组织中NE、DA等易化记忆过程递质的含量,同时,又降低了对记忆有阻抑作用的5-HT的含量,提示脑忆源片辅助改善学习记忆的作用可能与其改变脑组织内单胺类递质的含量有关。 Objective To investigate the effect of Naoyiyuan Tablets(NYYT) on contents of monoamine neurotransmitters, and to explore its mechanism for improving learning and memory abilities. Methods HPLC-ECD was used to detect the contents of monoamine neurotransmitters in cerebral homogenate of rats which were fed with NYYT for 15 days. A Phenomenex luna C18 column(150×4.60mm, 5 m) was used with a mobile phase of acetonitfile-200 mmol·L^-1 natrium biphosphoricum(13 : 87). The lion-rate was 1 mL·min^-1. The column temperature was 35 ℃. Results Monoamine neurotransmitters of NE, E, DA, 5-HIAA and 5-HT showed good linearity in the range of 0.0095-10.5 ng·μL^-1(r〉0.9996). The lowest detectable limit of E was below 5 ng·mL^-1, while NE, DA, 5-HIAA and 5-HT below 1.21 ng·mL^-1. Their average recoveries were in the range of 84.41%-100.92 %. Compared to the blank group, the levels of NE, E, DA were increased, but 5-HT was decreased in high-, middle-and low-dose NYYT groups, the changes of NE, DA and 5-HT contents being in a dose-effect manner. Conclusion The method is accurate, highly sensitive and easy to operate, and can be used to detect the contents of the monoamine neurotransmitters in rat cerebral homogenate. The mechanism of NYYT for improving learning and memory is probably related with the regulation of the contents of monoamine neurotransmitters.
