人参皂苷Re对大鼠心肌缺血再灌注细胞凋亡及Caspase-3的影响 被引量:18

摘要 目的:探讨人参皂苷Re对大鼠心肌缺血再灌注细胞凋亡及Caspase-3的影响。方法:采用健康大鼠30只,随机分为假手术组、模型组、人参皂苷Re治疗组,每组各10只,进行急性心肌梗死造模。Re治疗组结扎LAD前10min舌下静脉注射Re 30mg/kg。各组于再灌注6h后处死,检测细胞凋亡数目及Caspase-3酶活性。结果:人参皂苷Re治疗组细胞凋亡指数明显低于模型组(P<0.05)。Caspase-3酶活性测定及蛋白表达指数低于模型组(P<0.05)。结论:人参皂苷Re在减少急性缺血再灌注损伤所致的大鼠心肌细胞凋亡的同时,也减少Caspase-3的表达。以上结果提示人参皂苷Re抑制Caspase-3表达,可能是人参皂苷Re抑制急性缺血再灌注损伤诱导心肌细胞凋亡的机制之一。 Objective:To investigate the ginsenoside Re on myocardial ischemia-reperfusion apoptosis and Caspase-3 effect.Methods:30 healthy rats were randomly divided into sham operation group,model group,ginsenoside Re treated group,n = 10,modeling of acute myocardial infarction.Re the treatment group before LAD ligation 10min sublingual intravenous Re 30mg/kg.Each group were sacrificed after 6 hours reperfusion,the number of apoptotic cells were detected and Caspase-3 activity.Results:The treatment group ginsenoside Re apoptosis index was significantly lower than model group(P0.05).Determination of Caspase-3 activity and protein expression index lower than the model group(P0.05).Conclusion:Ginsenoside Re in reducing ischemic reperfusion injury induced myocardial apoptosis,but also reduce the expression of Caspase-3.These results suggested that inhibition of ginsenoside Re Caspase-3 expression,may be inhibited ginsenoside Re acute ischemia-reperfusion injury induced by one of the mechanisms of myocardial cell apoptosis.
