摘要
用人肺腺癌细胞株AGZ—83a常规免疫BALB/C小鼠,取脾细胞与小鼠骨髓瘤细胞SP2/0融合,获得一稳定分泌抗人肺腺癌单克隆抗体(McAb)的杂交癌细胞林──—QMC-WL99。制备并纯化了小鼠腹水抗体,腹水效价为5.5X106,亚型鉴定为IgG1。免疫组化结果显示QMC-WL99单抗的特异性较高,与肺腺癌有较好选择反应性,与正常组织无交及反应。该单克隆抗体的亲和常数为7.25×1010M-1,液氮冻再后复苏,杂交瘤细胞株QMC-WL99分泌抗体的能力不受影响。
Monoclonal antibody(McAb) QMC - WL99 were prepared by fusing SP2/0. cells with spleen cells of BALB/C mice immunized with the human lung adenocarcinoma cells AGZ - 83a. The titers of McAb in the ascites reached 5. 5 x 106. The immunoglobulin subclass of McAb QMC - WL99 was IgG1. Immunohistochemical results showed that McAb QMC- WL99 had a higher specificity and reacted with the lung adeno carcinoma tissue and not with normal tissues. The affinity constant of McAb QMC - WL99 was 7. 25 x 1010 M- 1. After repeated frozen storage and revival, the QMC - WL99 hybridoma cell line can steadily secret high titers of McAb.
