摘要
目的该研究探讨了2型糖尿病(T2DM)患者外周血单个核细胞(PBMC)组蛋白修饰情况。方法收集12例T2DM患者及12例健康对照者的PBMC,采用H3K4/H3K9甲基化检测试剂盒提取组蛋白和检测H3K4/H3K9甲基化水平。实时荧光定量PCR法检测T2DM患者的PBMC组蛋白甲基转移酶和组蛋白去甲基化酶基因的mRNA表达水平。结果与健康对照组比较,T2DM患者的PBMC组蛋白H3K4甲基化水平升高。实时定量PCR结果显示T2DM患者组PBMC的SET1、SUV39H1、SUV39H2、KDM3A、KDM5B和LSD1基因表达分别是健康对照组的3.06、0.46、0.48、0.43、3.07和0.35倍。结论 2型糖尿病患者外周血PBMC组蛋白甲基化呈现异常。
[ Objective ] To study file histone methylafion patterns in peripheral blood mononuelear cells (PBMC) from type 2 diabetic patients. [Methods] PBMC from 12 type 2 diabetic patients and 12 healthy controls were recruited. The global H3K4/H3K9 methylation was determined by assay kit. The differential expression of the histone methyltransferases and histone demethylases were measured by real-time polymerase chain reaction (RT-PCR). [Results] Increased global H3K4 methylation was observed in PBMC of type 2 diabetic patients compared with controls. The mRNA level of gene SET1, SUV39H1, SUV39H2, KDM3A, KDM5B, LSD1 in patients' PBMC compared with healthy controls was 3.06, 0.46, 0.48, 0.43, 3.07, 0.35 fold respectively. [Conclusion] Histone methylation appears abnormal in PBMC of Type 2 diabetic patients.
