摘要
目的研究GnRHa对子宫内膜异位症(简称内异症)患者离体培养异位、在位子宫内膜间质细胞生长抑制及分泌VEGF的影响。方法体外原代培养人内异症异位、在位子宫内膜间质细胞,用10-10M、10-8M和10-6M的GnRHa分别干预,用MTT法测定内膜间质细胞的生长增殖情况;用ELISA法测定其分泌的血管内皮生长因子(VEGF)浓度的变化。结果 GnRHa可抑制内异症异位及在位内膜间质细胞的生长增殖,呈剂量和时间依赖性(P<0.05),且对异位内膜间质细胞生长的抑制率明显高于在位(P<0.05);GnRHa可降低异位及在位内膜间质细胞分泌的VEGF的浓度,呈剂量依赖性(P<0.05),且高浓度(10-6M)对异位强于在位(P<0.05)。结论 GnRHa可明显抑制子宫内膜异位症患者异位、在位子宫内膜间质细胞的生长,降低其分泌VEGF的浓度。
[Objective] To investigate the effect of GnRHa on the cell proliferation and VEGF's secreting of ectopic and eutopic endometrial stromal cells from patients with endometriosis(EMs) in vitro. [ Methods ] The eetopic and eutopic endometrlal stromal cells were isolated, cultured and identified in vitro, then added to 10-10M, 10-8M, 10-6M GnRHa. The growth and proliferation of ectopic and eutopic stromal cells were measured by MTT assay; ELISA was used to measure the VEGF concentration change. [Results] GnRHa can inhibit the proliferation of ectopic and eutopic endometrial stromal cells from patients with endometriosis, in a dose-and time- dependent manner (P 〈0.05), and the effect on ectopie was stronger than the eutopie (P 〈0.05); The concentration of VEGF in ectopic and eutopic cells was significantly decreased after the treatment of GnRHa, in a dose-dependent manner(P 〈 0.05), and the ectopic was lower than the eutopic in lO-6M GnRHa (P 〈0.05). [Conclusion] GnRHa can significantly inhibit the cell proliferation of ectopic and eutopic endometrial stromal cells of EMs and decrease the secretion of VEGF.
