摘要
利用聚丙烯酰胺凝胶电泳( P A G E)和分光光度法,对添加不同激素组织培养的绞股蓝愈伤组织的过氧化物酶( P O D)同工酶及总皂甙含量进行了测定。结果表明:添加 N A A 培养的愈伤组织的 P O D 同工酶在阴极区酶带活性强,添加 B A 培养的愈伤组织的 P O D 同工酶在阳极区产生了新的酶带,同时添加 N A A、 B A、2.4- D 三种激素培养的愈伤组织的绞股蓝总皂甙含量最高。
By using the methods of PAGE and spectrophotometer,the peroxidase (POD) isozymes and the content of total gypenosides in Gynostemma callus in Vtro adding hifferent hormone were determined. The results showed that the POD isozymes activity rised in Gynostemma callus when methyl α naphthyl acetate (NAA) was added to the media; a new POD actvity pattern in Gynostemma callus arised when the media contained BA. Meanwhile, the highest content of total gypenosides in Gynostemma callus was obtained in the treatment of 3 kinds of hormones, NAA, 2, 4-D and BA.
