摘要
目的探讨抑癌方对小鼠结肠癌移植瘤微血管(microvessel,MV)生成和血管生成拟态(vasculogen-ic mimicry,VM)形成的影响。方法用CT26结肠癌细胞建立小鼠结肠癌移植瘤模型,分生理盐水(nor-mal saline,NS)组,抑癌方高、中、低剂量组及化疗药组,分别灌服NS,抑癌方高、中、低剂量及腹腔注射5-氟脲嘧啶(5-fluorouracil,5-Fu)。处理后比较各组之间瘤体质量;采用血管内皮细胞CD34免疫组织化学法和CD34免疫组织化学法+糖原蛋白过碘酸希夫氏双染法分别检测瘤组织中微血管密度(microvessel density,MVD)及血管生成拟态密度(vasculogenic mimicry density,VMD)。结果与NS组比较,抑癌方各剂量组及化疗组瘤体质量均显著降低(P<0.05);但抑癌方各组瘤体质量均显著高于化疗组(P<0.05)。NS组癌巢间MV和VM丰富,与之相比较,抑癌方各剂量组和化疗组瘤组织中MVD和VMD均显著减少(P<0.05)。结论抑癌方对小鼠移植瘤有一定的抑制作用,能够抑制结肠癌移植瘤中MV和VM的表达,这可能是抑癌方抗肠癌的作用机制之一。
Objective To research the effects of Yiaifang(YAF),a traditional Chinese medicine prescription,on microvessels(MV) and vasculogenic mimicry(VM) formation in mice bearing transplanted colon cancer.Methods The model of transplanted colon cancer was induced by colon cancer cell CT26 in mice.All mice were randomly divided into normal saline(NS) treated group,high-,medium-and low-dose YAF treated groups and chemotherapy group.Mice in NS treated group were fed with NS;while mice in the high-,medium-,low-dose YAF treated groups were fed with corresponding dose Yiaifang,respectively;and mice in the chemotherapy group were intraperitoneally injected with 5-fluorouracil.Tumor weights were observed and compared among all groups after the mice were executed using dislocation of cervical vertebra.The microvessel density(MVD) and vasculogenic mimicry density(VMD) in the tumor tissues of each group were assayed by using CD34 immunohistochemical staining of vascular endothelial cells,and double staining with CD34 immunohistochemistry and periodic acid-Schiff staining of glycoproteins,respectively.Results Tumor weights in high-,medium-and low-dose YAF treated groups and the chemotherapy group were significantly lower than those in NS treated group(P〈0.05);while tumor weights in high-,medium-,and low-dose YAF treated groups were significantly higher than those in chemotherapy group(P〈0.05).A plenty of MV and VM were found between the tumor nests in NS treated group,The levels of VMD and MVD in all dose YAF treated groups and chemotherapy group were significantly decreased compared with those in NS treated group(P〈0.05).Conclusion YAF has some effects on mouse transplanted tumor by inhibiting the formation of MV and VM,which may be one of anti-cancer mechanisms.
