局部化疗对宫颈鳞癌组织中Bcl-2、Bax表达水平及细胞凋亡的影响

摘要 目的探讨局部化疗前后宫颈癌组织中凋亡相关基因Bcl-2、Bax表达及细胞凋亡的变化及临床意义。方法应用免疫组织化学法检测接受及未接受局部化疗的宫颈癌组织中Bcl-2、Bax蛋白的表达,流式细胞仪检测细胞凋亡率。结果接受局部化疗的宫颈癌组织中,Bcl-2表达降低,bax表达升高,细胞凋亡率上升。卵巢组织中情况相同。结论局部化疗可能诱导了肿瘤细胞的凋亡,Bcl-2/Bax平衡的改变参与了凋亡调控,同时局部化疗对卵巢组织也有一定程度的影响,可能具有降低宫旁转移率、预防肿瘤细胞扩散的作用。 Objective To study the apoptosis and the expression of apoptosis-related gene bcl-2 and bax in the cervical carcinoma cells with and without local chemotherapy and to determine the mechanism of local chemotherapy and its clinical significance.Methods The expression of bcl-2 and bax were evaluated by immunohistochemistry in 40 specimens of cervical cancer and flow-cytometry was introduced in the study of apoptosis.Results In the tumors with and without local therapy,the expressions of bcl-2 was down.The expressions of bax was rise.The positive rates of apoptosis was up.It was same in the ovary.Conclusion The local therapy may induce the apoptosis of tumor cells,which also has influence on the ovary to depress parametrial metabasis and prevent the diffusion of the tumor cells.The changes of the balance between bcl-2 and bax may participate in the regulation of apoptosis.The expressions of Bcl-2 and Bax and the ratio of Bcl-2/Bax might be the evaluation index of the sensitivity of the local chemotherapy and the effect of therapy.
