摘要
目的:探讨重组人血管内皮抑制素恩度抑制异位内膜生长的作用机制,进而为子宫内膜异位症(EMS)的临床治疗提供理论依据。方法:将35只裸鼠随机分为正常组(n=10)和实验组(n=25)。实验组25只小鼠采用皮下种植方法建立子宫内膜异位裸鼠模型,造模成功的再随机分为对照组(n=11)和恩度组(n=11)。恩度组腹腔注射恩度2 mg.(kg.d)-1,对照组每日给予同等剂量的PBS液。用药2周后,观察小鼠血清细胞因子(VEGF、TNF-α)的水平及进行免疫组化检测异位内膜VEGF的表达。结果:对照组的细胞因子水平高于正常组和恩度组(P<0.05),且VEGF的表达高于恩度组。结论:恩度明显抑制VEGF等细胞因子水平,降低其在异位内膜组织中的表达,从而抑制异位内膜血管生长,达到抑制异位病灶的生长。
Objective: To evaluate mechanism of inhibition of ectopic endometrial growth with endostar,then to provide a theoretical basis for the clinical treatment of patients with EMS.Methods: 35 nude mice were randomly divided into normal group and experimental group,25 mice that were subcutaneously implanted in a methods used to establish nude mouse model of endometriosis.Then the models were randomly divided into control group and endostar group.Endostar group were given the endostar by intraperitoneal injection of 2 mg·(kg·d)-1,control group given the same daily dose of PBS solution.Two weeks later,serum were collected to detect serum cytokines(VEGF,TNF-α) levels and immunohistochemical methods were taken to determine the expression of VEGF.Results: The model group had higher levels of cytokines and the expression of VEGF than treatment group(P0.05).Conclusion: Endostar significantly inhibit levels of VEGF and other cytokines,reduce the expression of VEGF in the ectopic endometrial tissue,thus inhibiting the growth of ectopic endometrial blood vessels,and ectopic lesions.Thereby endostar can achieve the purpose of EMS treatment.
