无蹼壁虎抗肿瘤活性成分对ECV304细胞增殖和凋亡的影响 被引量:1

Effects of the Fresh GEKKO SWINHONIS Anti-neoplasm Active Component on the Proliferation and Apoptosis of ECV304 Cells

摘要 目的观察无蹼壁虎抗肿瘤活性成分对人脐静脉肉皮细胞系ECV304细胞增殖和凋亡的影响。方法常规培养ECV304细胞,设正常对照组、无蹼壁虎抗肿瘤活性成分高浓度(200mg/L)组、中浓度(20mg/L)组、低浓度(2mg/L)组。采用免疫细胞化学法检测不同实验组细胞PCNA的表达,并计算细胞阳性率为细胞增殖指数(PI);应用AO/EB荧光染色技术检测无蹼壁虎抗肿瘤活性成分对细胞凋亡的影响。结果①无蹼壁虎抗肿瘤活性成分高,中浓度组与正常对照组比较,ECV304细胞PCNA的表达降低,细胞增殖指数降低,差异有显著性(P〈0.01);②与正常对照组比较,无蹼壁虎抗肿瘤活性成分高,中高浓度组均能促进ECV304细胞凋亡。结论无蹼壁虎抗肿瘤活性戍分能够抑制ECV304细胞增殖,引起细胞凋亡。 Objective To investigate the effects of the fresh GEKKO SWINHONIS anti-neoplasm active component on the proliferation and apoptosis of human umbilicus vein endothehal cell hne ECV304 cells. Methods The ECV304 cells were cultured. Cells were randomly divided into normal control group,200mg/L,20mg/L,2mg/L of the fresh GEKKO SWINHONIS anti-neoplasm active component group. PCNA in ECV304 was determined by immunocytochemical method and the positive rate of cell was calculated. The cell apoptosis was measured by AO/EB fluorescent staining technique. Results (1)Compare with the normal control group,the expression of PCNA and PI of ECV304 cells were decreased in the 200mg/L and 20mg/L of the fresh GEKKO SWINHONIS anti-neoplasm active component groups; (2)The apoptosis of ECV304 cells were promoted in the 200mg/L and 20mg/L of the fresh GEKKO SWINHONIS anti-heoplasm active component groups. Conclusion The results suggest that the possible mechanism of the fresh GEKKO SWINHONIS anti-neoplasm active component in anti-tumor is to inhibit proliferation and apoptosis of the endothelial cell,then depress the visualization and suppress tumor growth at last. tosis
