丹参酮ⅡA磺酸钠注射液对小鼠Lewis肺癌生长及PCNA表达的影响

THE INHIBITORY EFFECTS OF THE GROWTH AND PCNA ESPRESSION ON MOUSE LUNG LEWIS CANCER BY TANSHINON IA

目的:观察丹参酮ⅡA磺酸钠注射液对小鼠Lewis肺癌的生长抑制及PCNA表达的影响,进而对丹参酮ⅡA的抗癌机制进行探讨。方法:建立Lewis肺癌小鼠模型,将动物随机分为生理盐水对照组、丹参酮ⅡA组、丹参酮ⅡA+环磷酰胺(CTX)组和环磷酰胺治疗有效组共四组。腹腔连续注射给药。10d后计算各组Lewis肺癌小鼠的瘤重、抑瘤率。用免疫组化方法测定PCNA表达水平。结果:与盐水组相比丹参酮ⅡA组及丹参酮ⅡA联合环磷酰胺组的瘤重明显减低、抑瘤率升高,对小鼠Lewis肺癌的生长有抑制作用,PCNA指数明显降低。结论:丹参酮ⅡA可抑制小鼠Lewis肺癌PCNA的异常表达并抑制小鼠Lewis肺癌生长。

Objective：To observe the inhibitory effect of TanshinoneⅡA sulfonic acid Injection of tumor growth and immune responses and PCNA expression on the mice with lewis lung cancer in order to explore the mechanism of its resisting-cancer function.Methods：To establish Lewis pulmonary carcinoma models in 40 C57BL/6 male,they mice were prepared and randomly divided into 4 groups（10 in each group）： sodium chloride（0.9%NS）,TanshinoneⅡA groups,TanshinoneⅡA with CTX group and the CTX therapy group.Each group was given with intraperitoneal injection.On 11th day the mice were killed.Then the tumor mass weight,the tumor growth inhibition rate were determined at the end of experiment,detected the expressions of PCNA by immunohistochemistry（IHC）.Results：TanshinoneⅡA groups,TanshinoneⅡA with CTX group were able to inhibit the growth of lewis lung cancer in the mice,the average weight of lewis lung cancer were reduced significantly.The tumor inhibitory rate were higher and PCNA was lower obviously. Conclusion：The TanshinoneⅡA injection can inhibit effectively the growth and metastasis of implanted Lewis lung carcinoma,which is probably attributed to reducing the abnormal expression of PCNA.
