摘要
目的探讨姜黄素对实验性糖尿病大鼠肾脏蛋白激酶C(PKC)由细胞浆到细胞膜移位活化的抑制作用。方法 36只大鼠随机分为正常对照组(A组)、糖尿病组(B组)及姜黄素治疗组(C组),每组12只。采用腹腔单剂量注射链脲佐菌素(STZ)65mg/kg建立糖尿病大鼠模型。C组给予姜黄素30mg·kg-1·d-1腹腔注射。实验第3周、第6周3组分别宰杀6只大鼠,并检测24h尿白蛋白排泄率(UAER)、肌酐清除率(Ccr)及肾重/体重;采用酶联免疫吸附法(ELISA)测定PKC活性。结果 B组UAER、Ccr、肾重/体重及肾脏细胞膜PKC活性均明显升高(P<0.05或<0.01),C组大鼠上述指标均明显下降(P<0.05或<0.01)。结论姜黄素通过抑制糖尿病大鼠肾脏细胞PKC移位活化而对糖尿病大鼠早期肾脏病变有保护作用。
Objective To investigate the inhibitory effect of curcumin on the spatiotemporal dynamics of PKC activation in the kidney of experimental STZ-induced diabetic rats.Methods 36 rats were randomly divided into three groups:normal control rats(group A),STZ-induced diabetic rats(group B),STZ-induced diabetic rats treated with curcumin 30mg·kg-1·d-1 intraperitoneally(group C),12 rats in each group.The UAER,Ccr and the profile of kidney hypertrophy were observed after 3,6 weeks of treatment,while the endogenous PKC activity at plasma membrane and in cytosol was measured by ELISA.In addition,the changes of renal tissues were observed by microscopy.Results The kidney/body weight ratio,Ccr,UAER,and PKC activity at plasma membrane in diabetic rats treated with curcumin were significantly lower than those in untreated diabetic rats(P0.05 or P0.01),meanwhile,the renal pathological changes in diabetic rats treated with curcumin were significantly improved.Conclusion Curcumin has some protective effects on experimental STZ-induced diabetic rats by inhibiting the spatiotemporal dynamics of PKC activation in the kidney.
