摘要
目的:探讨人体外周血CD4+CD25+细胞胞浆FoxP3与胞膜CD127的关系,为用CD4+CD25+CD127-作为界定调节性T细胞(Treg)的标志提供依据。方法:利用三色流式细胞计数分别测定32份外周血CD4+CD25+T细胞和CD4+T细胞中CD4+CD25+CD127-细胞和CD4+CD25+FoxP3+细胞百分率,对两者作相关性研究分析。结果:人体外周血CD4+CD25+T细胞和CD4+T细胞中CD4+CD25+CD127-与CD4+CD25+FoxP3+细胞的百分率均呈正相关(r值分别为0.962和0.803,P<0.01),CD4+CD25+CD127-的检测比CD4+CD25+FoxP3+检测更简便快捷。结论:人体外周血CD4+CD25+T细胞和血CD4+T细胞中CD4+CD25+CD127-与CD4+CD25+FoxP3+细胞百分率呈正相关,CD4+CD25+CD127-可以作为界定Treg细胞的标志及其检测。
Objective To investigate the association of membrane CD127 with intracellular FoxP3 of CD4^+CD25^+ in the human peripheral blood to provide the evidence of natural regulatory T cells(Tregs)-identified marker(CD4^+CD25^+CD127-).Methods The proportion of CD4^+CD25^+CD127-cells and CD4^+CD25^+FoxP3^+ cells in CD4^+CD25^+ T cells and CD4+ T cells from 32 patients were detected by 3 colors-flow cytometry and correlatively analyzed.Results The expression of membrane CD127 was negatively correlated with intracellular FoxP3^+ of CD4^+CD25^+ T cells in the peripheral blood.There was a positive correlation between the proportion of CD4^+CD25^+CD127-T cells and CD4+ CD25+ FoxP3^+T cells in CD4+ CD25+ T cells and CD4+ T cells(r value was 0.962 and 0.803 respectively,P less than 0.01).Detecting CD4^+CD25^+CD127-T cells showed the easier and faster advantages compared with the detecting CD4^+CD25^+FoxP3^+ T cells.Conclusion There was a positive correlation between the proportion of CD4^+CD25^+CD127-T cells and CD4^+CD25^+FoxP3^+ T cells in CD4^+CD25^+ T cells and CD4+ T cells.CD4^+CD25^+CD127-could be used to identify the Tregs and test it.
