洛伐他丁的电化学行为研究

摘要 用线性扫描极谱法考察了洛伐他丁的极谱行为,旨在建立一种简单、快捷和灵敏的测定洛伐他丁含量的分析方法。实验采用三电极系统(滴汞电极为工作电极,Pt电极为对电极,饱和甘汞电极为参比电极),以1∶1的0.8 mol/L NH4Cl和0.8 mol/L NH3.H2O混合溶液为底液,发现洛伐他丁在-1.62 V左右有一个灵敏的二阶导数极谱峰,其峰高与洛伐他丁溶液在0.50~7.00μg/mL范围内呈线性关系。洛伐他丁的加标样品回收率在98.0%~104.2%之间,最小检测限为0.370μg/mL。同时考察了表面活性剂和共存的金属离子Al3+、Cu2+、Fe3+、pb2+对洛伐他丁极谱波的影响。利用建立的洛伐他丁含量的极谱分析方法对样品中洛伐他丁含量进行了测定,结果表明所建立的洛伐他丁极谱分析方法快速、准确,具有一定实用价值。 A simple,rapid,and sensitive single-sweep polarographic method has been developed for the determination of the content of lovastatin.In this experiment,the polarographic behavior of lovastatin was studied using three-electrode-system(DME as the working electrode,SEC as the reference electrode,and Pt wire served as the counter electrode).By using the mixture of 0.8mol/L NH3· H2O and 0.8mol/L NH4Cl with volume ratio 1∶1 as supporting electrolyte,a sensitive polarographic wave of lovastatin was detected at about-1.62V(SCE).The results show that the height of derivative polarographic wave peak is linearly correlated with the concentration of lovastatin in the range from 0.50 to 7.00μg/mL,the coefficient of correlation is 0.9989,the recovery for the sample was 98.0%~104.2%,and the minimum detection limit was 0.370μg/mL.Under the optimum conditions of experiment,the influences of surface active agent and some metal ions,including Al3+,Cu2+,Fe3+ and Pb2+,on the polarographic wave of lovastatin were studied,the results show that the metal ions and surface active agent affect the height and position of the polarographic wave peak to different degrees.According to Ep=-1.62V of lovastatin,the content of lovastatin in sample was determined,and the results show that the polarographic analysis method for the single-sweep polarograpy behavior study of sudan dyes is rapid,accurate,and feasible.
