摘要
目的:了解不同取样拭子对沙眼衣原体抗原检测结果的影响。方法:4个稀释度的沙眼衣原体培养物,分别用2种国产拭子和5种进口拭子吸取等量的培养物,进行ELISA法检测,比较不同拭子对测定结果的影响。结果:以无拭子阳性对照为标准,棉拭子(拭子1、2)、涤纶拭子(拭子3、4)和人造丝拭子(拭子5)与阳性对照的吸光度差异有统计学意义(P〈0.05)。尼龙植绒拭子(拭子6、7)与无拭子阳性对照的测定结果一致,显著高于棉拭子、涤纶拭子和人造丝拭子(P〈0.05)。7种拭子空白对照全部为阴性,但女性棉拭子空白的吸光度最高,与阴性对照、尼龙植绒拭子、男性棉拭子、涤纶拭子和人造丝拭子空白差异有统计学意义(P〈0.05)。样本释放均一性方面,尼龙植绒拭子的吸光度变异系数较小,低于男性棉拭子和男性涤纶拭子。吸水性方面,棉拭子和人造丝拭子需要3~5秒吸收培养物,植绒拭子需要30~40秒,涤纶拭子需要60~90秒。结论:尼龙植绒拭子优于涤纶拭子和人造丝拭子,棉拭子会导致不同程度的漏检。拭子的选择是分析前质量控制的环节,提高检验质量要重视分析前质量控制。
Objective:7 varieties of swabs have been investigated to assess their effectiveness in the detection of chlamydia trachomatis. Methods:Three clinical strains of Chlamydia trachomatis serovar E,F and J were serially passaged in McCoy cell in flat-bottomed culture plates.The Chlamydia were harvested and prepared as samples for the detection.Two kinds of swabs made in China and the other five kinds of swabs made in foreign country were choosed,.Chlamydia trachomatis antigen was detected by Enzyme-linked immunosorbent assay.The results were analysed by SPSS 13.0.Results:Among all of the detection,Dacron(swab1,2),cotton(swab3,4) and rayon(swab5) swabs have showed statistically significant compared with controls(P0.05).Nylon flocked swabs(swab6,7)have agreement results with controls.The absorbance of nylon flocked swabs were higher than other swabs and have showed statistically significant compared with Dacron,cotton and rayon swabs.The absorbance of blank swabs were negative for 7 kinds of swabs.The absorbance of female cotton swab was higher than the other swabs.The coefficient of variation for nylon flocked swabs was low.Cotton and rayon absorbed the culture immediately.However,Dacron need 60~90 seconds and nylon flocked need 30~40 seconds.Conclusion:Nylon flocked swabs were better than cotton,rayon and Dacron swabs for the detection of chlamydia trachomatis.Choosing suitable swabs is the way to improve quality control.
