人脑胶质瘤中CEACAM1和CD105的表达及意义 被引量:3

摘要 目的探讨癌胚抗原相关细胞黏附分子1(carcinoembryonic antigen-related cell adhesion molecule1,CEACAM1)、CD105在人脑胶质瘤中的表达及其与微血管生成的关系。方法应用免疫组织化学方法,检测62例人脑胶质瘤及15例正常脑组织中CEACAM1、CD105的表达情况,并记录微血管密度(microvessel density,MVD),对脑胶质瘤及正常脑组织中CEACAM1、CD105的表达情况进行相关性分析。结果①Ⅰ-Ⅳ级胶质瘤组织CEACAM1标记指数分别为(15.78±4.93)%、(19.64±7.31)%、(38.05±8.57)%和(51.07±8.44)%,随病理分级升高逐渐增高(r=0.857,P<0.01),与正常脑组织比均有统计学差异(P<0.01)。②正常脑组织CD105蛋白表达阴性,而各级胶质瘤组织均有CD105蛋白阳性表达。高倍视野(×100)下Ⅰ-Ⅳ级胶质瘤组织CD105标染的微血管密度(CD105-MVD)分别为9.97±2.36、13.03±3.16、16.17±3.21和20.13±2.72,随病理分级提高逐渐增高(r=0.780,P<0.01),且不同级别胶质瘤间差异有统计学意义(P<0.01)。③CEACAM1标记指数与CD105-MVD呈显著正相关(r=0.889,P<0.01)。结论 CEACAM1在胶质瘤中的表达增加可能引起胶质瘤的发生、发展;CD105标记的肿瘤内微血管密度(MVD)对评估胶质瘤恶性程度有重要意义;CEACAM1可能作为一种促血管生长因子而刺激肿瘤血管的生成。 Objective To investigate the relationship between the expression of carcinoembryonic antigen-related cell adhesion molecule 1(CEACAM1) ,CD105 and angiogenesis in human glioma. Methods Immunohistochemical method was used to analyze the expression of CEACAM1 and CD105,and calculate the MVD marked with CD105 in 62 specimens of human glioma and 15 specimens of normal human brain tissue. Results CEACAM1 labeling index(CEACAM1 LI) were(15.78±4.93) %,(19.64±7.31) %,(38.05±8.57) % and(51.07±8.44) % in the glioma of grades Ⅰ-Ⅳ,respectively.The CEACAM1 LI in the gliomas was significantly higher than that in the normal brain tissue(P0.01) .The CEACAM1 LI was positively related with the grading of gliomas(r=0.857,P0.01) .The expression of CD105 was negative in all the normal brain tissues,but positive in all the glioma tissues.The microvessel densities immunostained by CD105(CD105-MVD) under high-power field(×100) in the glioma of grades Ⅰ-Ⅳ were 9.97±2.36,13.03±3.16,16.17±3.21 and 20.13±2.72 respectively(P0.01) .The CD105 was positively related with the grading of gliomas(r=0.780,P0.01) .CEACAM1 LI was positively correlated to CD105-MVD in the glioma(r=0.889,P0.01) . Conclusion Overexpression of CEACAM 1 may induce glioma development.CEACAM1 may stimulate tumor angiogenesis as a vascular growth factor.Microvessel density(MVD) marked with CD105 may be an important marker for evaluating the malignancy of glioma.
