摘要
利用赤麂SRY基因序列设计引物,通过PCR反应分别扩增出毛冠鹿、黑麂、小麂的SRY序列,并测得630 bp的碱基序列。麂属动物之间的序列差异为0.79%-1.90%,毛冠鹿属与麂属之间的序列差异为3.02%-3.97%。以牛SRY为外源,用NJ法和MP法进行毛冠鹿和麂属3种动物分子系统进化树的构建与分析,结果表明:麂属3种动物之间的分歧时间为79万-190万年,毛冠鹿与麂属动物间的分歧时间为302万-397万年,梅花鹿与麂属动物的分歧时间为349万-460万年。
According to the SRY gene of Indian muntjac,the SRY genes of tufted deer(Elaphodus cephalophus),black muntjac and Chinese muntjac were amplified by PCR.Fragments of SRY gene(each 630 bp in length) were characterized by direct sequencing of PCR products.The nuclearted divergences of the SRY region were 0.79%-1.90% among three species of Muntiacus,3.02%-3.97% between the tufted deer and three species of Muntiacus.The phylogenetic tree was contructed and analyzed by NJ method and MP method.The results of phylogeny relationship showed that divergence time was about 79-190 Myr among three species of Muntiacus,the tufted deer diverged from Muntiacus about 302-397 Myr ago,and Cervus Nippon diverged from Muntiacus about 349-460 Myr ago.The tufted deer was a transitionary type in the deer.
