姜黄素对人胃癌SGC-7901细胞增殖及COX-2mRNA和COX-2蛋白表达的影响 被引量:5

摘要 目的:探讨姜黄素对人胃癌SGC-7901细胞增殖及对环氧酶-2(C0X-2)基因及蛋白表达的影响。方法:应用MTT法考察浓度姜黄素对胃癌SGC-7901细胞增殖的影响;应用RT-PCR、Western-blotting法检测COX-2 mRNA及蛋白的表达。结果:5~80μg.mL-1姜黄素对胃癌SGC-7901细胞增殖的抑制作用呈剂量依赖性和时间依赖性。姜黄素对COX-2mRNA和COX-2蛋白的表达有不同程度的抑制作用,随着剂量的增加,抑制作用增强,较高浓度(20~80μg.mL-1)的姜黄素对COX-2 mRNA和COX-2蛋白的表达有显著抑制作用(P<0.01)。结论:姜黄素可能通过抑制COX-2 mRNA和COX-2蛋白的表达抑制胃腺癌细胞SGC-7901的增殖,可能成为一种胃癌预防剂。 OBJECTIVE:To investigate the effects of curcumin on the proliferation of gastric cancer cell line SGC-7901 and the gene /protein expressions of cyclooxygenase-2(COX-2).METHODS:The effect of different concentration of curcumin on the proliferation of gastric cancer cell line SGC-7901 was determined by MTT method.The expressionsof COX-2 mRNA and COX-2 protein were detected by reverse transcriptase polymerase chain reaction(RT-PCR)andWestern blot assay,respectively.RESULTS:The inhibitory effect of curcumin(5-80μg·mL-1)on the proliferation ofgastric cancer cell line SGC-7901 was in a dose-and time-dependent manner.Curcumin showed different degree ofinhibitory effects on the expressions of COX-2 mRNA and COX-2 protein in a dose-dependent manner;at a highconcentration(20-80μg·mL-1),curcumin showed significant inhibitory action on the expressions of COX-2 mRNA and COX-2 protein(P0.01).CONCLUSION:Curcumin can inhibit the proliferation of SGC-7901 through down-regulation on the expressions of COX-2 mRNA and COX-2 protein,which is expected to become a preventative agentfor gastric cancer.
