摘要
目的:建立一种新型的体外培养成骨细胞的生物学模型。并对其生物学特性进行研究。方法:选择大耳白兔骨髓基质细胞为材料进行体外成骨细胞培养实验,在不同培养体系中进行体外培养,通过倒置显微镜观察、透射电镜、扫描电镜、组织化学染色技术、钙染色等手段对获得的细胞进行生物学特性进行研究。结果:获得的细胞呈多种形态,有长短不一、大小不均的胞浆突起,且互相连接,细胞互相重叠呈复层生长,并形成细胞结节;胞浆丰富,富含线粒体、粗面内质网、高尔基体等,具有合成分泌功能旺盛的结构,与典型成骨细胞的形态结构相似。同时,胞浆富含碱性磷酸酶活性,在条件培养基中细胞发生钙化现象,这与典型成骨细胞的生物学特性相似。且获得的细胞在体外能连续传代,形态和功能不变。结论:我们认为用骨髓基质细胞为材料进行体外培养是成骨细胞体外培养的一种新型的生物学模型,为人工骨替代材料的研制。
Objective: To establish a new model of osteoblasts culture in vitro and study it's biological characterization.Methods: The stromal cell of bone marrow was obtain from the trochanteric region of femur of rabbit and was cultured in the different cultural fluid dddin vitro. the cells were examined by phase contrast microscope, transmission electron microscope, scanning electron microscope, histochemistry stains and calvaria strains. Results:The stromal cells were increased and lined up in one layer, had the same morphological feature. ALP activity, and demineralized matrix formed in vitro to those of osteoblasts. Conclusion:The cultrued stromal cells derived from the bone marrow were a new model of osteoblast culur in vitro.
