高体积分数氧暴露早产儿外周血单个核细胞血红素加氧酶-1表达水平及其活性的变化 被引量:1

Changes of Expression and Activity of Heme Oxygenase-1 in Peripheral Blood Mononuclear Cell of Hypero-xia-Exposed Premature Infants

摘要 目的观察高体积分数氧(高氧)暴露早产儿血单个核细胞(PBMC)血红素加氧酶-1(HO-1)蛋白及mRNA的表达水平和活性变化。方法分别用反转录聚合酶链反应(RT-PCR)技术、Westernblot方法对60例吸入高氧的早产儿(高氧组)血中PBMC中HO-1mRNA及蛋白表达水平进行检测,并测定血清胆红素水平代表HO-1活性及血浆一氧化碳血红蛋白(COHb)百分比,并与吸入常压空气的早产儿60例对照进行比较。结果与空气组相比,高氧组早产儿PBMC中HO-1mRNA及蛋白表达明显增加[(0.64±0.05)vs(0.13±0.03),(1.78±0.25)vs(1.12±0.31),Pa<0.01)],血清胆红素生成量及血浆COHb水平也显著升高[(76.25±11.33)nmol.L-1.h-1vs(53.47±8.73)nmol.L-1.h-1,(6.73±0.42)μmol.L-1vs(2.47±0.34)μmol.L-1,Pa<0.01]。结论高氧暴露可上调早产儿血PBMC中HO-1mRNA及蛋白质表达水平,推测HO-1可能参与早产儿高氧肺损伤。 Objective To investigate the changes of expression and activity of heme oxygenase-1(HO-1)protein and mRNA in peripheral blood mononuclear cell(PBMC)of hyperoxia-exposed premature infants.Methods The expression of HO-1 mRNA and protein in the PBMC of 60 cases of hyperoxia-exposed preterm infants(hyperoxia group)and 60 cases of air-exposed preterm infants(air group)were measured by reverse transcription polymerase chain reaction(RT-PCR)and Western blot,respectivesly,and the bilirubin in serum and carboxy hemoglobin(COHb)in plasma were determined also.Results Compared with air group,the expressions of HO-1 mRNA,HO-1 protein,the bilirubin and COHb in hyperoxia group significantly increased[(0.64±0.05)vs(0.13±0.03),(1.78±0.25)vs(1.12±0.31),(76.25±11.33)nmol·L-1·h-1 vs(53.47±8.73)nmol·L-1·h-1,(6.73±0.42)μmol·L-1 vs(2.47±0.34)μmol·L-1,Pa0.01].Conclusion Hyperoxia-exposure upregulates the expressions of HO-1 and HO-1 protein,and HO-1 may be associated with the hyperoxia-induced lung injury of premature infants.
