纳洛酮对催产素所致小鼠记忆减退的影响

摘要 目的观察侧脑室分别给予纳洛酮、催产素(oxytocin,OT)对小鼠学习记忆的影响及阿片受体在OT影响小鼠学习记忆中的作用。方法按分层随即区组设计将小鼠分成4组(n=7):生理盐水组(NS组)、纳洛酮组(Nal组)、OT组、Nal+OT组。采用避暗实验分别测定给药后第1天、第2天小鼠的潜伏期及错误次数。结果用药后第2天,与NS组相比,OT组错误次数增加(P〈0.01),潜伏期缩短(P〈0.05);与OT组相比:Nal+OT组的错误次数减少(P〈0.05),潜伏期差异无统计学意义(P〉0.05)。结论在本实验条件下,侧脑室微量注射OT能损伤小鼠的记忆功能,而侧脑室给予阿片受体拮抗剂纳洛酮后,可逆转OT所致的记忆损伤,提示OT损害记忆的作用与阿片受体有关。 Objective To investigate the effect of naloxone or oxytocin (OT) microinjection into lateral cerebral ventricle on the memory in mice, and to explore the role of opioid receptor in the effect of OT on memory in mice. Meth- ods The mice were randomly divided into 4 groups ( n = 7 each) : normal saline group ( NS group ), naloxone (Nal) group, OT group, Nal + OT group. The times of the errors and the duration of latency were respectively recorded on the first day and the second day after the drug administration in step - through task. Results On the second day, as com- pared to the NS group, the times of errors increased (P 〈 0.01 ) and the duration of latency decreased (P 〈 0.05 ) in the step - through task in the OT group ; Compared with the OT group, the times of errors decreased ( P 〈 0.05) while there were no significant differences in the duration of latency ( P 〉 0.05 ) in the step - through task in the Nal + OT group. Conclusion Under this experiment conditions, microinjection of OT lateral cerebral ventricle could impair the memory in mice. Meanwhile, naloxone, the opioid receptor antagonist, could reverse the effect of OT. therefore, the opioid receptor mediated the effects of OT on the memory.
