阿霉素-多孔磷酸三钙陶瓷缓释体的研制及体内释药试验 被引量:5

摘要 目的:研制阿霉素多孔磷酸三钙陶瓷( A P T C P)缓释体并为骨肿瘤保肢治疗术后骨缺损的修复提供新方法。材料与方法:制备 P T C P陶瓷,将阿霉素载入其中制得 A P T C P。实验动物分成 A、 B两组, A 组右股骨粗隆中植入 A P T C P, B组静脉注射阿霉素。不同时期取粗隆部位的肌肉及血浆标本,用高效液相色谱法测定肌肉及血浆中阿霉素的浓度。结果:体内植入 A P T C P后,阿霉素能在局部缓慢释放达10 周以上,局部组织药浓高,全身药浓低。既能缓释阿霉素控制肿瘤复发,又能充填并修复骨缺损。结论: A P T C P缓释体,可望成为骨肿瘤保肢治疗术后骨缺损修复的一种新方法。 Objective:To manufacture adriamycin porous tricalcium phosphate ceramics(A PTCP)drug delivery system(DDS)as a possible method for bone defects after bone tumor operation.Methods:A PTCP DDS were made by putting adriamycin into PTCP.Rabbits were divided into two groups(A and B).A PTCP were implanted in great trochanters of right femora in Group A.Adriamycin was injected into veins in Group B.Muscle around A PTCP and plasma were taken out at different periods.Adriamycin concentrations in muscle and plasma were measured by high performance liquid chromatography(HPLC).Results:A PTCP could release gradually over 10 weeks.Adriamycin concentrations in the muscle were higher than that in plasma in GroupA. A PTCP DDS could release gradually adriamycin to prevent bone tumor recurrence and repair bone defects after bone tumor operation.Conclusion:A PTCP may be a new method for repairing bone defects after bone tumor operation.
