气郁证模型大鼠的病理特征 被引量:3

Pathyologic Status of Qi-Stagnation Model Rats

摘要 目的探讨气郁证的病理基础。方法将20只健康雄性Wistar大鼠随机分为对照组与气郁组,采用慢性束缚法建立气郁证大鼠模型,观察气郁证大鼠的精神、体重、摄食量、蔗糖水摄取量,检测血清皮质酮(CORT)、血浆血管紧张素(Ang)、内皮素(ET),血清5羟色胺(5-HT)、去甲肾上腺素(NE)、肾上腺素(E)、血清一氧化氮(NO),计算胸腺及肾上腺指数,透射电镜观察胸主动脉组织内皮细胞超微结构的变化。结果气郁组大鼠表现为精神萎靡,反应迟钝,摄食量减少,体重增长缓慢;蔗糖水摄取量、摄食量及体重均明显低于对照组(P<0.01);气郁组大鼠血浆ET、Ang、血清CORT、NE、E升高,NO、5-HT降低(P<0.05或P<0.01),胸腺、脾脏指数明显降低(P<0.05);胸主动脉内皮细胞超微结构发生明显改变。结论气郁证大鼠存在明显的血管内皮结构和功能的损害,神经、内分泌、免疫系统紊乱失衡是气郁证的病理基础。 Objective To explore the pathyologic basis of qi stagnation. Methods Twenty healthy male Wistar rats were randomized into normal group and model group. The rat models with Qi stagnation were established by chronic constraint for 6 weeks. The general condition including spirit,weight,food and sucrose water intake of the models were recorded. The level of serum cortisone (CORT),plasma angiotensin Ⅱ (AngⅡ),endothelin (ET),serum 5-hydroxy-trytamine (5-HT),norepinephrine (NE),epinephrine (E),and nitric oxide (NO) were measured. The thymus and adrenal gland indices were calculated. Changes in ultrastructure of thoracic aorta endothelial cells were observed with transmission electron microscope. Results Compared with the normal group,the model rats showed listlessness,slow response,reduction of diet,and weight increase becoming slower,food intake,sucrose water intake,and weight were significantly reduced (P〈0.01). The level of plasma ET,AngⅡ,serum CORT,NE,and E of the model group increased significantly,and the level of NO and 5-HT decreased (P〈0.05 or P〈0.01). The thymus and spleen indices were significantly lowered (P〈0.05),and the ultrastructure of thoracic aortic endothelial cells was obvious changed. Conclusion The model rats with Qi stagnation have obvious endothelial damage in the structure and function. Their neural-endocrine-immune system disorder serves as the pathyological basis of qi stagnation.
