摘要
目的探讨至生胶囊在体内、体外对食管癌Eca-109细胞的作用及作用机理。方法体内实验:将BALB/c裸鼠50只随机分为至生胶囊低、中、高剂量组、环磷酰胺注射液(CTX)组、空白组5组,每组10只。造模成功后,低、中、高剂量组分别灌服至生胶囊0.25g/kg、0.5g/kg、0.75g/kg;CTX组按40mg/kg给CTX;空白组给等容积生理盐水,各组均每日1次,连续10天,观察各组小鼠体重及瘤重的变化。体外实验:将日本大耳白兔10只随机分为至生胶囊低、中、高剂量组、CTX组及空白组5组,每组2只,制备含药血清,分别加入Eca-109细胞悬液中,培养72h,测OD值,显微镜下观察细胞的形态学及细胞数目改变。结果体内实验:至生胶囊低、中、高剂量减轻瘤重的作用随浓度的增加而增强(P<0.05),呈一定的量效关系,至生胶囊中剂量组在抑制肿瘤的同时能增加小鼠体重。体外实验:至生胶囊高剂量组对Eca-109细胞的抑制作用与CTX相当(P>0.05);至生胶囊对Eca-109细胞的抑制作用呈明显的量效关系(P<0.05);镜下观察,各至生胶囊组及CTX组见部分细胞出现皱缩、变小变圆、折光性降低、脱壁死亡等。结论至生胶囊在体内、体外实验中,均对人食管癌Eca-109细胞有明显的抑制作用,呈量效关系,其抗肿瘤作用可能与引起肿瘤细胞凋亡有关。
Objective To observe the inhibition of the Zhisheng Capsule (To-life Capsule) in vivo and in vitro on human esophageal cancer Eca-109 cells and to study its functional mechanism. Methods In vivo:Fifty BALB/c mice were randomized into five groups,i.e. Zhisheng Capsule small,medium,large dosage group,cytoxan (CTX) group and blank control group,10 mice for each group. After medeling,Zhishen Capsule small,medium and large dosage group were given Zhisheng Capsule 0.25g/kg,0.5g/kg and 0.75g/kg respectively,CTX group was given CTX 40mg/kg,and blank control group was given normal saline. All groups were treated once per day for 10 days to observe changes of body weight and tumor weight. In vitro:Ten Japanese white rabbits were equally divided into samll,medium,and large dose of Zhisheng Capsule groups,CTX group,and blank control group. The medicine-contained serum was added into the Eca-109 cells suspension. After cultured for 72 hours,the optical density (OD) value was measured. The microscopic observation on the changes in morphology and number of cells were carried out. Results The effect of Zhisheng Capsule on tumor weight was stronger with the increase of concentration (P〈0.05),showing dosage-effect relation,body weight was increased accompained with the inhibition of tumor weight in Zhisheng Capsule medium dose group. The inhibition effect on Eca-109 cell was equal in Zhisheng Capsule large dose group and CTX group (P〉0.05). Zhisheng Capsule significantly inhibited Eca-109 cells with dose-effect rebation (P〈0.05). The microscopic observation showed that some cells became shrunken,changed to smaller in size and round in shape,decreased in refraction,and died of off-wall in all treated groups. Conclusion Zhisheng Capsule significantly inhibits human esophageal cancer Eca-109 cells with a dose-effect relationship. Its anti-tumor effect may be related to the cause of apoptosis of tumor cells.
