摘要
目的:研究香港远志黄酮类成分对H2O2损伤PC12细胞的保护作用。方法:应用现代分离鉴定技术分离香港远志中黄酮类成分;96孔板培养PC12细胞24h加入100μmol的受试样品(以维生素C为对照),培养24h后加入400μmolH2O2作用2h。使用MTT法检测各组细胞活性,用自动酶标仪在570nm波长下测定各孔吸光度。结果:香港远志中黄酮类成分山柰酚-3-O-芸香糖苷、山柰酚-3-O-葡萄糖苷、芒果苷、香港远志黄酮A、香港远志黄酮B具有一定的对H2O2损伤PC12细胞保护作用的活性。结论:本实验可为防治阿尔茨海默病新药的研发提供理论基础。
OBJECTIVE: To study the protective effect of flavonoids from Polygala hongkongensis on H2O2 induced PC12 cells damage. METHODS: Flavonoids of P. hongkongensis was acquired by modem isolation and identification methods; PC12 cells were cultured in 96 well plates for 24 h. Then 100 μmol of tested samples (vitamin C as control) were added. After 24 h, cells were treated with 400 μmol H2O2, for 2 h. MTT assay was used to detect activity of PC12 cell. The absorption of each well was measured by automatic reader at 570 rim. RESULTS: Kaempferol-3-O-rutinoside, kaempferol-3-O-glucoside, mangiferin, HongKong polyhongkongenoxanthone A and polyhongkongenoxanthone B from P. hongkongensis had protective effect on H2O2 induced PC12 cell damage. CONCLUSION: The study can be used as research theoretic basis of new drugs for alzheimer's disease.
