摘要
目的:优选金银花中绿原酸微波辅助提取工艺,并对提取物的抗氧化能力进行分析。方法:以甲醇浓度、料液比、提取时间为考察因素,以提取液在327nm波长的吸光度和二苯代苦味酰基自由基(DPPH·)清除率为响应值,采用响应曲面法优选工艺;通过比较提取液对DPPH·的清除能力分析提取物的抗氧化能力。结果:最佳提取工艺为以43%的甲醇溶液作提取剂、按液料比40mL·g-1、在微波功率400W下提取8min,绿原酸提取率为7.62%;金银花提取物对DPPH·的清除能力与绿原酸含量呈正相关。结论:优化的工艺稳定、合理、可行,提取率高、提取时间短;绿原酸在金银花提取物的抗氧化能力中起主要作用。
OBJECTIVE: To optimize the microwave-assisted extraction process of chlorogenic acid from Lonicerare japonica and to analyze the anti-oxidation function of the extract. METHODS: The extraction technology of chlorogenic acid was optimized by response surface methodology using the concentration of methanol, ratio of material to liquid and extraction time as factors and taking the absorbance of extract at 327 nm and the scavenging rate of extract for free radical as response. The anti-oxidation function of the extract was analyzed by comparing the scavenging capacity of extract against DPPH .. RESULTS: The optimal extraction process was as follows: 43 % menthol as solvent, the ratio of material to liquid was 40 ml·g-1, extracting for 8 min. The field of chlorogenic acid was 7.62% The scavenging capacity of extract for DPPH. was direct correlation with the content of chlorogenic acid. CONCLUSION: The optimized extraction process is stable, reasonable and feasible with high field and short extraction time. Chlorogenic acid plays a major role in the anti-oxidation of extract of L. japonica.
