摘要
目的测定蝙蝠葛酚性碱片灌胃后大鼠血浆中蝙蝠葛碱的质量浓度。方法以原阿片碱为内标,采用C18柱,乙腈-体积分数4%乙酸-三乙胺(体积比80∶240∶2)为流动相,流速为1.0 mL.min-1,检测波长282 nm,柱温30℃。结果蝙蝠葛碱的线性范围为0.05~5.0 mg.L-1(r=0.997 9),定量下限为50μg.L-1,日内、日间精密度均小于15%。结论本方法专属性强、灵敏度高、回收率高、重现性好,可用于蝙蝠葛碱血药质量浓度测定及药动学研究。
Objective To investigate the method for determination of dauricine in rat plasma after ig. administration of phenolic alkaloids of Menispermum dauricum tablets. Methods Protopine was used as the internal standard. The samples were separated on a Diamonsil C18 column(4. 6 mm×200 mm,5 μm)at 30℃. The mobile phase consisted of acetonitrile-4% acetic acid and triethyl amine(V: V: V = 80: 240:2 ). The flow rate was 1.0 mL·min^-1 and the UV absorbance detection was set at 282 nm. Results The calibration curve of dauricine was linear in the concentration range of 0. 05- 5.0 mg·L^-1 ( r = 0. 997 9 ). The lower limit of quantification was 50 μg·L^-1. The RSDs of inter-and intra-day were less than 15%. Conclusions A sensitive,precise and accurate method for the determination of dauricine in plasma is described, which can be used in its pharmacokinetic studies.
