摘要
通过克隆技术及体内移植,从自建的成骨肉瘤细胞 L T H O S(简称 S)中获得两株细胞克隆 S1 和 S2,应用组织形态学观察、染色体分析、软琼脂克隆形成法以及裸小鼠体内成瘤试验研究癌细胞的生物学特性。结果发现 S1 和 S2 两株均保持人类染色体核型,所形成肿瘤显示人成骨肉瘤上皮样组织形态,其中 S1 的细胞电泳率高达1.0, S2 为0.70, S1 的群体倍增时间为22.4 h,软琼脂形成率为33.5% , S2 群体倍增时间为24.0 h,软琼脂形成率为23.1% 。裸小鼠右后肢皮下接种 S1 和 S2,发现 S1 成瘤时间短,细胞增殖快,但在4 周内两者均不发生转移;通过尾静脉注射 S1 和 S2 细胞,发现4 周时,接种 S1 细胞的裸鼠出现广泛的实验性肺转移,接种 S2 细胞的裸鼠肺部仅见几个分散的癌细胞。
In order to study the characteristics of two cell lines (S 1,S 2) derived from human osteosarcoma cell LTHOS(S) and established by clone technique and transplantation.Cell growth in soft agar,chromosome analysis,histological observation,and tumor formation rate in nude mice were used to characterize the cells.Electrophoresis mobility was 1.00 and 0.70,the doubling time of population was 22.4 h and 24.0 h,the clone formation rate in soft agar was 33.5% and 23.1% for S 1 and S 2,respectively.Extensive lung tumor was observed in nude mice after inoculation of S 1 cells through tail vein.The results indicated that the cell lines isolated may be very useful material for the study of characteristics of metastatic cancer cells.$$$$
