鸡传染性支气管炎病毒纤突蛋白的目的基因点突变对其免疫原性的影响

EFFECT OF SITE MUTATION IN SPIKE 1 PROTEIN GENE ON IMMUNOGENICITY OF THE AVIAN INFECTIOUS BRONCHITIS VIRUS

反转录聚合酶链反应（ Ｒ Ｔ－ Ｐ Ｃ Ｒ） 扩增鸡传染性支气管炎病毒（ Ｉ Ｂ Ｖ） 中国流行株（ Ｈ Ｄ）的主要免疫原纤突蛋白 Ｓ１ 基因（ Ｓｐｉｋｅ ｐｒｏｔｅｉｎ ｇｅｎｅ） ，将其插入载体ｐ Ｕ Ｃ１８ 的 Ｂａｍ ＨⅠ／ Ｈｉｎｄ Ⅲ位点，在大肠杆菌中实现目的基因的分子克隆。经克隆化 Ｓ１ 基因的限制性酶切片段多态性（ Ｒ Ｆ Ｌ Ｐ）分析和 Ｓｏｕｔｈｅｒｎ 杂交之后，双脱氧链终止法测定其５′端高变区核苷酸序列，并以此与 Ｇｅｎｅ Ｂａｎｋ 中的参考毒株 Ｍａｓｓａｃｈｕｓｓｅｔｔｓ４１ 相应序列作比较，分析其同源性。在此基础上，分别以此两毒株抗原研制灭活疫苗，交叉免疫攻毒。结果表明，从 Ｓ１ 基因５′端高变区的起始密码子 Ａ Ｔ Ｇ 至第３５０ 位，两毒株仅在起始密码子后第１３１ 位存在一个碱基的点突变，由 Ｍ４１ 的 Ｃ 突变为 Ｈ Ｄ 株的 Ｇ，即 Ｓ１蛋白上的第４１ 个氨基酸由 Ｍ４１ 的 Ａｌａ 突变为 Ｈ Ｄ 株的 Ｇｌｙ ，二者核苷酸同源率为９９７ ％ ，氨基酸同源率为９９１ ％ 。两毒株灭活抗原疫苗免疫后的交叉攻毒试验表明， Ｍ４１ 毒株疫苗对 Ｈ Ｄ 株的保护率为７５ ％ ，而 Ｈ Ｄ 株疫苗对 Ｍ４１ 株的保护率只有６５ ％ 。

After cloning and identification of the gene encoding spike l protein of the avian infectious bronchitis virus(IBV) HD isolate of China, the sequence of the 5' end within the hypervariable region (HVR) of the S1 gene was analyzed, and compared with the corresponding sequence of the IBV standard M41 strain from the GeneBank. One bp site mutation on the analyzed sequence was found between M41 and the HD strains, which was located at the 350nt after ATG, i. e. the Gly in HD replaced for Ala in M41 at the 41 amino acid residue of S1 glycoprotein. Cross protection between two vaccine strains showed that significant different protective effect can be induced by the site mutation on 5' end within the HRV of the S1 gene on the immunogenicity of IBV.
