二苯乙烯苷对Aβ_(1-42)诱导的PC12细胞损伤的保护作用

The protective effects of stilbene glycoside on the PC12 cells injury induced by Aβ_(1-42)

目的观察二苯乙烯苷(TSG)对Aβ1-42诱导损伤的PC12细胞生长、分化的保护作用。方法实验分为对照组、Aβ1-42组和TSG高(10μmol/L)、中(1μmol/L)、低(0.1μmol/L)剂量组,测定乳酸脱氢酶(LDH)漏出量,采用硫代巴比妥酸比色法测定丙二醛(MDA),黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)活性,噻唑兰(MTT)法检测细胞活力,免疫组化检测bcl-2表达。结果 TSG能明显减少Aβ1-42引起细胞损伤。TSG处理后能明显提高细胞活力,降低LDH漏出和细胞凋亡率。结论 TSG能明显减少Aβ1-42对PC12细胞损伤,可能与抗氧化、抗自由基和脂质过氧化有关。

Objective To study the protective effects of stilbene glycoside（TSG） on the PC12 cells injury induced by Aβ1-42.Methods All cells were divided into 3 groups： one group serving as the saline control group（control group）,one group receiving oxidative damage PC12 cells model was induced by Aβ1-42（Aβ1-42 group）,one group receiving oxidative damage PC12 cells model was induced by Aβ1-42 after given TSG 2 h at TSG 10,1,0.1 μmol/L once.The survival rate of the cells was determined by MTT,the content of LDH was determined by ultraviolet spectrophotometry and the content of malonaldehyde（MDA） and SOD activity were measured respectively by thiobarbituric aicd and xanthine oxidese methods.The expression of bcl-2 was observed by immunohistochemistry.Results TSG reduced obviously cells injury induced by Aβ1-42.10 μmol/L TSG improved the cells survival rate,reduced LDH releasing and MDA content,increased SOD activity.Conclusions TSG has significantly protective effect on the PC12 cells injury induced by Aβ1-42.
