摘要
目的观察解聚复肾宁(JJFSN)对高糖诱导的大鼠肾小球系膜细胞(GMCs)表达血小板衍生生长因子-B(PDGF-B)的影响。方法制备厄贝沙坦(Irb)和不同剂量JJFSN含药血清,加入到高糖诱导的体外培养的GMCs中。采用ELISA检测不同时间(24、48、72 h)细胞培养上清液中PDGF-B含量,分别采用RT-PCR法、W estern印迹检测48 h细胞PDGF-B mRNA及其蛋白表达。结果与正常对照组比较,高糖组PDGF-B分泌增加、PDGF-B mRNA及其蛋白表达上调(P<0.001)。与高糖组比较,Irb和JJFSN两种含药血清均能逆转上述变化,后者的抑制作用强于前者(P<0.05),且抑制作用呈一定的时间、剂量依赖关系。结论高糖可致GMCs分泌PDGF-B增加、PDGF-B mRNA及其蛋白表达上调,JJFSN可减少PDGF-B分泌,下调PDGF-B mRNA及其蛋白表达,从而有利于延缓糖尿病肾病肾小球硬化(GS)的进展。
Objective To investigate the effect of jiejufushenning(JJFSN) on the secretion,expressions of platelet-derived growth factor-B(PDGF-B) in rat glomerular mesangial cells(GMCs) cultivated under high glucose.Methods The two drug-contained serum of model rats were prepared,then GMCs were cultured with high concentration glucose(HG) and HG plus irbesartan,different dose of JJFSN for different duration(24,48,72 h),and set up normal control simultaneously.The content of platelet-derived growth factor-B(PDGF-B) in the supernatant of cultured GMCs were detected by ELISA,and the expressions of PDGF-B were detected by RT-PCR,Western Blot,respectively.Results Compared with normal control group,the content of secretion of PDGF-B in the supernatant of cultured GMCs was higher in HG(P0.001),and the expression of PDGF-B was up-regulated in HG group(P0.001).Compared with HG group,HG plus irbesartan,and high,middle,low dose of JJFSN significantly decreased PDGF-B secretion in the supernatant of cultured GMCs(P0.005),and decreased the expressions of PDGF-B,JJFSN groups were more significant(P0.05),a certain dose-dependent and time-dependent was shown.Conclusions GMCs can increase secretion and expressions of PDGF-B induced by high glucose,JJFSN can reduce the secretion of PDGF-B,and down-regulate the expressions of PDGF-B,then delay the progression of glomerular sclerosis of diabetic nephropathy.
