HPV感染的宫颈病变对血清锌、IL-2、IL-10及组织E2F1表达的影响 被引量:7

Significance and Expression of Serum Zn,IL-2,IL-10 and E2F1 in Patients with High-Risk HPV Infection of Cervical Lesions

摘要 目的:研究高危型人乳头状瘤病毒(HPV)感染的宫颈病变患者血清锌、白细胞介素(IL)-2、IL-10的含量和组织中E2F1的表达及其临床意义。方法:选取宫颈上皮内瘤样病变(CIN)Ⅰ级组、CINⅡ级组各30例,CINⅢ级及宫颈癌组各20例,均为高危型HPV感染。慢性宫颈炎患者30例(对照组),HPV阴性。分别用原子吸收光谱法测定血清锌,酶联免疫吸附法(ELISA)测定IL-2及IL-10水平,免疫组化法测定宫颈组织中E2F1表达。结果(:1)血清锌、IL-2的含量在CINⅡ级组、CINⅢ级组、宫颈癌组中逐级降低,3组间比较差异均有统计学意义,且3组分别与CINⅠ级、对照组比较差异亦均有统计学意义(均P<0.01)。宫颈癌组血清IL-10高于CIN各组,CINⅢ级组高于Ⅱ级组,差异均有统计学意义(P<0.05或P<0.01)。组织中E2F1的表达在宫颈癌组、CIN组及对照组间比较差异有统计学意义(P<0.005),CIN各组内相比差异无统计学意义(P>0.005)。(2)血清IL-2与锌水平呈正相关(P<0.05)。血清IL-2与IL-10水平、组织E2F1表达均呈负相关(P<0.05)。结论:高级别宫颈病变及宫颈癌患者中存在Th1/Th2的漂移,可能影响了机体对HPV的清除。机体缺锌可能参与了这种变化。 Objective:To investigate the clinical significance of serum levels of zinc(Zn),interleukin(IL)-2,IL-10 and E2F1 in patients with the high-risk HPV infection of cervical lesion. Methods: The patients with cervical diseases were selected including 30 cervical intraepithelial neoplasia (CIN) Ⅰ, 30 CIN Ⅱ, 20 CIN Ⅲ and 20 cervical carcinoma with high-risk HPV infection, and 30 chronic cervicitis with HPV-negative infection. The serum level of Zn was detected by atomic absorption spectrometry. The values of IL-2 and IL-10 were detected by ELISA. The expression of E2F1 was detected by immunohistochemistry. Results: The serum levels of Zn and IL-2 were significantly decreased in the CIN Ⅱ, CIN Ⅲ and cervical cancer group (P 0.05). The level of IL-10 was significantly higher in cervical cancer group than that of CIN groups (P 0.05). The expressions of E2F1 were significantly different between cervical cancer group,CIN group and control group (P 0.05), but no significant difference between CIN groups (P 0.005). There was a positive correlation between the serum IL-2 and Zn (P 0.05). But the negative correlation was found between the serum IL-2, IL-10 and E2F1 (P 0.05). Conclusion: The drift of Thl to Th2 was found in patients of the high-clinical-level cervical disease and cervical cancer, which may affect the body’s clearance of HPV. Zinc deficiency may be involved in this change.
