枸杞多糖抑制对硫磷致血管内皮功能损伤的研究 被引量:1

摘要 目的:探讨枸杞多糖(LBP)抑制对硫磷(DNTP)所致的血管内皮功能损伤的作用。方法:DNHP与大鼠离体胸主动脉血管环(EVAPVR)和培养的人脐静脉内皮细胞(HUVEC)共孵30min,以LBP为保护、以超氧化物歧化酶(SOD)为对照,检测血管内皮依赖性舒张反应(EDRR)、内皮细胞单层通透性(ECMP)及细胞培养液生化指标。结果:血管环实验结果显示,LBP(10g/L)明显减轻了DNTP(4.5μmol/L)对血管EDRR的抑制作用;细胞实验结果显示,LBP(10g/L)明显降低了ECMP,增加了细胞培养液中SOD和一氧化氮(NO)的含量、降低了丙二醛(MDA)的含量(均P<0.05),与SOD对照组的作用相当。结论:LBP可抑制DNTP所致的血管内皮功能损伤,其机制可能与氧化应激有关。 Objective: To explore the mechanism of inhibitory effect of lycium barbarum polysaccharide (LBP) on damnification of blood vessel endothelium function induced by parathion (DNTP). Methods: The isolated rat thoracic aortic rings (EVAPOR) and cultured human umbilical vein endothelial cells (HUVEC) were incubated with DNTP for 30 min. LBP was used for protective effect and the level of superoxide dismutase (SOD) was used as the control. The endothelial-dependent relaxation reaction (EDRR), endothelial cell monolayer permeability (ECMP) and biochemical index were detected. Results: The result of the vascular circle experiment showed that LBP (10 g/L) obviously reduced the inhibitory effect of DNTP(4.5 μmol/L) on vascular EDRR. The result of cell experiment showed that LBP (10 g/L) obviously reduced the ECMP and MDA, and increased the level of SOD and nitric oxide (P 0.05, respectively). These effects were corresponded with the SOD control. Conclusion: LBP inhibited the damnifivation of blood vessel endothelium function induced by DNHP, which may be related to the oxidative stress.
