摘要
目的研究TPX2在不同级别脑胶质瘤组织中的表达,及其表达水平同胶质瘤病人生存时间的关系。方法正常脑组织、低级别和高级别脑胶质瘤标本分别为5、21、31例,采用免疫组化染色和Western blot法检测TPX2蛋白的表达,实时荧光定量PCR检测TPX2 mRNA的表达水平。结果 正常脑胶质细胞TPX2蛋白染色为阴性,绝大部分胶质瘤标本中可见TPX2蛋白的阳性表达,胶质瘤组织TPX2表达水平与病人生存时间成负相关(P<0.01)。TPX2蛋白及mRNA表达水平在胶质瘤组织中均明显上调,与正常脑组织相比差别显著(P<0.01),其在高级别与低级别胶质瘤中的表达有显著差异(P<0.05)。并且TPX2蛋白的相对表达量与胶质瘤的病理级别呈正相关(r=0.408,P<0.05)。结论 TPX2的表达可能同恶性胶质瘤进展有关。
Objective To investigate the expression level of TPX2 in different grade gliomas and the relationship between the expression of TPX2 and survival time of patients with gliomas.Methods There were 5,21 and 31 cases in normal brain tissues,low-grade and high-grade glioma specimens respectively.The expression of protein TPX2 was detected by immunohistochemistry and Western blotting and TPX2 mRNA by real-time PCR.Results TPX2 protein staining was negative in the normal tissues,while positive staining of TPX2 was found in most glioma specimens.There was a negative correlation between the expression level of TPX2 and life time of patient with glioma(P0.01).The expression levels of TPX2 protein and TPX2 mRNA were obviously increased in glioma tissues,and there was a significant difference between glioma tissues and normal brain tissue(P0.01),and between low-grade and high-grade glioma tissues(P0.05).In addition,there was a positive correlation between the expression of TPX2 and pathological grade of gliomas(r=0.408,P0.05).Conclusions The expression of TPX2 is associated with the progression of glioma.
