碘乙酰胺诱导成纤维细胞水通道蛋白1的表达具有能量依赖性 被引量:2

摘要 目的旨在观察细胞的能量代谢状态是否会影响水通道蛋白1(AQP1)的表达。方法采用体外培养的Balb/c 3T3小鼠成纤维细胞株,用肌酸激酶抑制剂碘乙酰胺(IA)作用于该细胞,应用免疫印迹和免疫组化技术检测细胞在0、4、6 h时AQP1的表达。结果细胞与IA作用4 h后,AQP1表达开始增加,至6 h时AQP1表达大量增加,同时伴有细胞死亡;RT-PCR发现AQP1 mRNA显著增加,干扰线粒体功能的微管抑制剂及呼吸链细胞色素c氧化酶阻断剂同样诱导了AQP1的表达。结论 IA诱导Balb/c 3T3小鼠成纤维细胞AQP1的大量表达存在着转录前的调节,并且具有线粒体相关的能量依赖性。 Objective To investigate the impact of energy metabolism at the cellular level on the expression of the water channel protein aquaporin 1 (AQP1). Methods Balb/c mouse fibroblasts were incubated with iodoacetamide (IA) in vitro, and the changes in AQP1 expression were detected by immunoblotting and immunohistochemistry at 0, 4, and 6 h. Results IA induced the expression of AQP1 at 4 and 6 h accompanied with cell death. Reverse transcription PCR showed an increased expression of AQP1 mRNA in the cells.
