摘要 目的探讨还原型谷胱甘肽(GSH)和γ-谷氨酰半胱氨酸合成酶(γ-GCS)在狼疮性肾炎(LN)病程中的作用。方法测定MRL/lpr狼疮鼠及正常对照组小鼠肾组织GSH和丙二醛(MDA)活性,荧光定量PCR和Western blot检测肾组织γ-GCS的表达。结果狼疮鼠肾组织中GSH浓度较对照组降低,MDA水平明显增高。狼疮鼠肾组织γ-GCS mRNA和蛋白表达均较对照组显著减少(P<0.05)。结论 MRL/lpr小鼠肾脏中存在氧化应激异常,γ-GCS表达异常降低从而导致GSH生成减少,机体抗氧化能力减低,更加剧了肾组织氧化损害。 Objective To investigate the role of glutathione (GSH) and γ-glutamylcysteine synthetase (γ-GCS) in lupus nephritis. Methods Spectrophotometry was used to measure the oxidative/anti-oxidative indices including malonyldialdehyed (MDA) and GSH in the kidney of MRL/lpr lupus mice. Quantitative PCR and Western blotting were used to detected the expression of γ-GCS.
