TNFα-和MMP-9在鼻息肉嗜酸粒细胞浸润过程中的作用 被引量:3

Effect of TNF-α and MMP-9 in the infiltration of eosinophile granulocyte in nasal polyps

摘要 目的:研究鼻息肉组织中TNF-α、MMP-9的表达和意义,并探讨两者表达与嗜酸粒细胞(EOS)浸润的关系。方法:采用原位杂交和免疫组织化学技术,检测鼻息肉组30例息肉组织及对照组10例下鼻甲黏膜组织中TNFα-、MMP-9 mRNA和蛋白的表达;采用苏木精-伊红染色观察鼻息肉和下鼻甲黏膜组织中EOS的浸润情况,并比较分析三者之间的相关性。结果:鼻息肉组织中TNFα-表达的阳性细胞数、阳性血管数及MMP-9阳性细胞数与下鼻甲黏膜组织相比差异有统计学意义(P<0.05);且鼻息肉组织中TNF-α阳性细胞数和阳性血管数均与EOS数存在明显相关性(P<0.05),MMP-9阳性细胞数与EOS数明显相关(P<0.05),TNF-α阳性细胞数与MMP-9阳性细胞数存在相关性(P<0.05)。结论:TNF-α、MMP-9在鼻息肉的发病机制中发挥着重要作用,TNF-α可能通过诱导MMP-9的产生促进EOS在鼻息肉中的浸润。 Objective:To study the expression and significance of TNF-α,MMP-9 and their relationship with the infiltration of eosinophile granulocyte in nasal polyps.Method:The expression of TNF-α and MMP-9 was determined in tissues of nasal polyps from 30 patients(nasal polyps group) and in inferior turbinate mucosa tissues from 10 patients(control group) by in situ hybridization and immunohistochemical technique,and the number of eosinophile granulocyte was counted in the same tissue by HE staining.Their correlations with each other were also analyzed in the tissue of nasal polyps.Result:The number of TNF-α and MMP-9 positive cells and TNF-α positive blood vessels in nasal polyps were more than that in control group(P〈0.05).The number of both TNF-α positive cells and blood vessels had positive relationships with the number of eosinophile granulocyte,but there was only positive relationship between the number of MMP-9 positive cells and eosinophile granulocyte(P〈0.05).At the same time there was a positive relationship between the number of TNF-α and MMP-9 positive cells(P〈0.05).Conclusion:TNF-α and MMP-9 may play an important role in the pathological mechanism of nasal polyps.TNF-α may induce the expression of MMP-9 and promote the migration of eosinophile granulocyte.
