巴西橡胶树DNA甲基化的MSAP分析 被引量:12

摘要 本文采用"甲基化敏感扩增多态性"(Methylation Sensitive Amplification Polymorphism,MSAP)的分析方法,在全基因组水平检测巴西橡胶树DNA甲基化位点,确定巴西橡胶树基因组DNA甲基化模式和水平。结果表明:16对MSAP引物选择性扩增,共扩增262条带,其中甲基化位点87个,甲基化比例为33.21%,其中,半甲基化位点为32个,比例为12.22%;全甲基化位点为55个,比例为20.99%。对部分巴西橡胶树基因组甲基化位点进行回收,最终分离了18条存在甲基化的基因组DNA序列。BLAST比对后,同源分析表明巴西橡胶树基因组中包括转录因子、蛋白激酶等在内的多种类型的DNA序列中均存在甲基化现象。本研究为深入研究巴西橡胶树的DNA甲基化奠定基础。 In this study,We detected rubber's DNA methylated sites and determined its patterns and levels at the whole genome level by Methylation Sensitive Amplification Polymorphism(MSAP).The results confirmed that 87 methylated sites were detected in 262 total sites by using 16 pairs of selective primers,which ratio of methylation was 33.21%.Of which hemi-methylated sites were 32 and methylation ratio was 12.22%.Of which full-methylation sites were 55 and methylation ratio was 20.99%.We get 18 sequences from extracted DNA methylated sites.After analyzed with BLAST,the homology search showed that DNA methylation of rubber existed in different types of DNA sequence such as Transcription Factor,Protein Kinase and so on.It paved the way for further research of DNA methylation of rubber.
