糜酶抑制剂对实验大鼠肺纤维化的干预作用

Effects of chymase inhibitor on bleomycin-induced pulmonary fibrosis in rats

摘要 目的观察用糜酶抑制剂对博莱霉素致大鼠肺纤维化模型干预作用。方法健康SD大鼠60只随机均分为正常对照组(生理盐水)、博莱霉素模型组和干预组(博莱霉素+糜酶抑制剂)。分别在第7、14、21和28天处死。HE染色观察支气管肺组织病理改变,免疫组化观察肺组织中Ⅰ型、Ⅲ型胶原的表达。结果干预组肺泡炎程度明显轻于模型组和对照组(P<0.05)。干预组肺纤维化程度比模型组明显减轻(P<0.05)。与对照组比较,干预组和模型组肺组织中Ⅰ型、Ⅲ型胶原均表达持续增强,但干预组明显减少(P<0.05)。结论糜酶抑制剂能减轻博莱霉素诱导的大鼠肺纤维化,可能是通过下调Ⅰ型、Ⅲ型胶原的表达。 Objective To study the effects of chymase inhibitor on bleomycin-induced pulmonary fibrosis in rats.Methods Sixty SD rats were randomly divided into three groups with 20 rats each.After the pulmonary fibrosis model was established by intratracheally instilling bleomycin 5mg/kg,rats in group A were given chymase inhibitor 10mg/kg by daily stomach instillation and those in group B were not treated.Rats in group C were given normal saline as the controls.Five rats of each group were killed on the 7th,14th,21st and 28th day,respectively,for observing pathological changes of lung tissue by HE staining and detecting the expressions of collagen and collagen Ⅲ in bronchial lung tisusues.Results Alveolitis was significantly less in groups of C and A than that in group B.The severity of pulmonary fibrosis was much lower in group A than that in group B.Compared to those in group C,the expressions of collagen and collagen Ⅲ in the lung fibrosis were higher in groups of B and A,which were much lower in group A than those in group B.Conclusion Chymase inhibitor can effectively alleviate bleomycin-induced pulmonary fibrosis in rats,possibly through inhibiting the expressions of collagen and collagen Ⅲ in lung tissues.
