重组白细胞介素13诱导成纤维细胞分泌巨噬细胞炎症蛋白1α

The expression of macrophage inflammatory protein-1α (MIP-1α) in 3T3 fibroblasts induced by recombinant interleukin13

摘要 目的观察重组白细胞介素13对3T3成纤维细胞巨噬细胞炎症蛋白-1αmRNA表达的影响。方法分别用DMEM(对照组)和40ng/ml、80ng/ml重组白细胞介素13(干预组)培养液作用于3T3成纤维细胞,24h和48h后MTT法观察细胞生长情况,用细胞增殖率表示;两组细胞24h后原位杂交方法检测巨噬细胞炎症蛋白-1αmRNA的表达。结果重组白细胞介素13可显著促进3T3成纤维细胞增殖,且呈时间-剂量效应关系(P<0.01)。对照组及重组白细胞介素13组均可检测到巨噬细胞炎症蛋白-1αmRNA的表达,但与对照组相比,重组白细胞介素13显著促进巨噬细胞炎症蛋白-1αmRNA的表达。结论重组白细胞介素13促进成纤维细胞分泌巨噬细胞炎症蛋白-1α,可能在肺纤维化的发生、发展过程中起重要作用。 Objective To investigate the effect of recombinant interleukin13(rIL-13) on mRNA expression of macrophage inflammatory protein-1α (MIP-1α) in 3T3 fibroblasts. Methods 3T3 fibroblasts were cultured in DMEM medium as control group and medium supplemented with 40ng/ml and 80ng/ml of rIL-13 for 24h and 48h respectively. MTT assay was carried out to study cell proliferation, the levels of MIP-1αmRNA in 3T3 fibroblasts were detected by hybridization. Results rIL-13 significantly increased fibroblast proliferation in a time-and dose-dependent manner ( 0.01). The expression of MIP-1αmRNA can be detected in the fibroblasts of both control group and rIL-13 groups, but the expression levels of MIP-1αmRNA in rIL-13 groups increased compared with control group( 0.01) . Conclusions rIL-13 can increase 3T3 fibroblast proliferation and significantly induce the expression of MIP-1αmRNA.
