摘要
本文探讨了电泳系统上槽泄露引起双向电泳凝胶蛋白点纵向拖尾问题.上槽泄露导致上槽SDS损耗,电流增大以及局部温度升高从而导致双向电泳凝胶高分子量区域蛋白点纵向拖尾.上槽泄露与上槽设计存在不足,使用者不适当操作以及上槽维护不良有关.对Ettan DALT six电泳系统而言,一旦出现上槽泄漏问题,建议更换改良后的新版上槽.电泳过程中应避免上槽泄露,SDS的不足或损耗,电流过大和电泳液温度过高.
This paper intended to solve the problem of vertical streaking caused by the leaking of upper buffer chamber(UBC) in two-dimensional gel electrophoresis(2-DE).UBC leakage resulted in sodium dodecyl sulfate(SDS) depletion,increased current and elevasted local temperature of the upper buffer,leading to vertical streaking in the high-molecular-weight region of 2-DE gels.The leaking problems could be caused by either the defected design of the 2-DE system or the improper usage /maintenance during operation.An improved Ettan UBC is recommended if the leaking is observed with an existing system to avoid the causes of unsatisfactory separation of electrophoresis.Meanwhile,the operation and maintenance following the manufacturers instructions is also very important.
