摘要
目的观察中药血必净注射液对脓毒症大鼠肝组织凝溶胶蛋白(GSN)及炎症因子的影响。方法雄性Wistar大鼠78只,随机分为正常对照组(n=6)、假手术组(n=24)、盲肠结扎穿孔术(CLP)组(n=24)、血必净组(n=24)。后两组采用CLP制备脓毒症大鼠模型。假手术组麻醉后开腹翻动盲肠后关腹。血必净组于术后2、12、24、36、48、60h静脉注射血必净注射液,每次剂量均为4ml/kg;假手术组及CLP组静脉注射同体积生理盐水。采用ELISA方法检测术后6、12、24、72h肝组织GSN、高迁移率族蛋白B1(HMGB1)的含量,并应用鲎试验检测肝组织中内毒素(LPS)的含量。结果与正常对照组及假手术组比较,CLP组大鼠肝组织GSN含量明显降低(P<0.05),而LPS及HMGB1水平明显升高(P<0.05)。与CLP组比较,给予血必净注射液治疗后,肝组织GSN含量显著升高(P<0.05),而LPS及HMGB1水平均明显降低(P<0.05)。相关分析显示,各时间点CLP组肝组织GSN含量与LPS水平呈显著负相关(P<0.05),与HMBG1水平无明显相关性(P>0.05)。结论血必净注射液可能通过拮抗内毒素的作用而促进肝组织GSN生成,进而减轻脓毒症的病理过程。
Objective To observe the effects of XUEBIJING injection on the expression of hepatic gelsolin(GSN) and inflammatory cytokine in septic rats.Methods Seventy-eight male Wistar rats were involved in present study and randomly divided into normal control group(n=6),sham operation group(n=24),cecal ligation puncture(CLP) group(n=24) and XUEBIJING group(n=24).The CLP sepsis models of rats were established with the animals in the latter two groups.XUEBIJING injection was injected intravenously at 2,12,24,36,48 and 60 hours after operation(4ml/kg each) in XUEBIJING group,while rats in sham operation group and CLP groups received same amount of normal saline by intravenous injection.The levels of hepatic GSN and high mobility group box-1(HMGB1) protein at 6,12,24 and 72 hours after operation were determined by ELISA,and limulus test was performed to measure the hepatic LPS levels.Results Compared with normal control and sham operation group,The hepatic GSN levels decreased and LPS and HMGB1 levels increased markedly in CLP groups(P〈0.05).Compared with CLP group,the hepatic GSN levels increased and LPS and HMGB1 levels decreased markedly in XUEBIJING group(P〈0.05).Correlation analysis revealed that a significant negative correlation existed between the hepatic GSN contents and LPS levels at each of time points in CLP group(P〈0.05),while no obvious correlation was found between GSN and HMGB1 levels(P〉0.05).Conclusion XUEBIJING injection may enhance the production of hepatic GSN via inhibition of LPS in septic rats,thereby having beneficial effect on septic response.
