摘要
目的探讨载脂蛋白E基因在转基因鼠体内的表达及其在血脂代谢中的作用。方法显微注射法建立H-ApoE转基因鼠,Southern杂交对阳性小鼠基因组鉴定。ELISA测定血清载脂蛋白E含量。酶法分别测定小鼠血清甘油三酯(TG)和血清胆固醇(TC)。结果转基因鼠血清载脂蛋白E含量分别为:tg4鼠(2.06±0.22)mg/L,tg7鼠(1.89±0.33)mg/L。血清TG水平转基因鼠〔12月龄:tg4(2.89±0.31)mmol/L,tg7(1.99±0.45)mmol/L〕与对照组鼠〔12月龄:(2.05±0.12)mmol/L〕比较有显著性差异(P<0.05),血清TC水平转基因鼠〔12月龄:tg4(3.02±0.14)mmol/L〕与对照组鼠〔12月龄:(2.11±0.01)mmol/L〕比较也表现出显著性差异(P<0.01)。结论 H-ApoE基因异常表达影响到了小鼠的血脂代谢。
Objective To establish transgenic mice and investigate the role of h-apoE gene in lipoprotein metabolism.Methods The human apoE transgenic mice were generated via microinjection.Southern blot hybridization was used to identify the expression of the gene.The human apoE level was measured in the serum of h-apoE transgenic mice with ELISA methods.The serum triglyceride and cholesterol were also elevated with the enzymatic methods.Results The human apoE gene could be expressed in the first generation of transgenic mice.The apo-E levels of transgenic mice(tg4(2.06±0.22) mg/L,tg7(1.89±0.33) mg/L) were significant.The serum triglyceride level were high in transgenic mice(12-month age groups: tg4(2.89±0.31)mmol/L,tg7(1.99±0.45)mmol/L) than that in the respective control mice(12月龄:(2.05±0.12)mmol/L)(all P0.05).And serum cholesterol level in the 12-month transgenic mice(tg4(3.02±0.14)mmol/L) were higher than that in control mice(2.11±0.01)mmol/L(P0.01).Conclusions The overexpression of apoE gene impairs the lipoprotein metabolism.
