摘要
目的:观察鸡尾酒疗法对大鼠急性脊髓损伤(acute spinal cord injury,ASCI)后细胞色素C、caspase-3表达和神经细胞凋亡的影响。方法:36只Wistar大鼠,被随机分为假手术组(A组)、生理盐水(normal saline,NS)20μl对照组(B组)、甲强龙(solu-medrol)30 mg/kg治疗组(C组)、鸡尾酒疗法20μl治疗组(D组)、鸡尾酒疗法10μl治疗组(E组)、鸡尾酒疗法5μl治疗组(F组),后五组统称为ASCI组,每组6只。采用压迫法建立大鼠急性脊髓损伤动物模型,ASCI组于伤后10 min进行药物治疗。建模成功后8 h取材,HE染色观察组织病理变化;免疫组织化学染色检测细胞色素C和caspase-3的表达,并进行统计学分析。结果:ASCI后8 h,HE染色结果显示鸡尾酒疗法治疗组脊髓组织病理学改变明显轻于其他损伤组;免疫组化染色表明,ASCI组细胞色素C和caspase-3阳性细胞率,D组明显小于B、C、E和F组(P<0.01)。结论:采用鸡尾酒疗法经蛛网膜下腔注射给药可有效减少急性脊髓损伤后细胞色素C、caspase-3的表达以及继发性神经细胞凋亡的发生,对损伤处临近节段的脊髓具有保护作用。
Objective:To investigate into the effect of cocktail therapy on the expression of cytochrome C,caspase-3 and neurocyte apoptosis after acute spinal cord injury(ASCI) in rats.Methods:A total of thirty-six Wistar rats were randomly assigned to 6 groups:sham-operated group(A group),saline control group(B group),Solu-Medrol therapy group(C group),cocktail therapy 20 μl-group(D group),cocktail therapy 10 μl-group(E group),and cocktail therapy 5 μl-group(F group).The ASCI model was introduced using the compression method;administration was conducted 10 min post-injury.Morphological changes were observed by the HE staining,and the expression of cytochrome C,caspase-3 was detected by immunohistochemical staining.Results:HE and immunohistochemical staining showed that cocktail therapy could significantly improve pathological changes and curtail the amounts of cytochrome C,caspase-3 positive cells(P0.01).Conclusion:Cocktail therapy by intrathecal interventions may effectively inhibit the expression of cytochrome C,caspase-3 and curtail neurocyte apoptosis after ASCI,exerting neuroprotection.
